Cyclophilin A Inhibitors Suppress Proliferation and Induce Apoptosis of MKN45 Gastric Cancer Stem-like Cells by Regulating CypA/CD147-Mediated Signaling Pathway

Hee Jeong Cho et al.

Summary: This study found that compound 9 and cyclosporin A are natural inhibitors of cyclophilin A (CypA), which can effectively suppress the growth of gastric cancer stem cells (GCSCs) by inducing cell cycle arrest, promoting apoptosis, and regulating AKT and MAPK signaling pathways through the inhibition of CypA/CD147 axis.

Discovery of a New CaMKII-Targeted Synthetic Lethal Therapy against Glioblastoma Stem-like Cells

Jang Mi Han et al.

Summary: The poor prognoses of patients with glioblastoma multiforme (GBM) are attributed to glioblastoma stem-like cells (GSCs) which induce drug resistance and rapid tumor recurrence. This study aims to explore a novel synthetic lethal therapy targeting calcium/calmodulin-dependent protein kinase II (CaMKII) for combatting GSCs. Through high-throughput drug combination screening, the combined treatment of CaMKII inhibitors and neurokinin 1 receptor (NK1R) inhibitors is found to exhibit chemical synthetic lethal effects on GSCs both in vitro and in vivo. It is proposed that this CaMKII-targeted combination therapy could be effective for treating GBM.

Lung Cancer Stem Cell Markers as Therapeutic Targets: An Update on Signaling Pathways and Therapies

Yue Zheng et al.

Summary: Cancer stem cells play a crucial role in tumor metastasis, relapse, and therapy resistance. The accuracy and biological function of these cells are still controversial. Biomarkers of cancer stem cells are used for cancer diagnosis, targeted therapy, and prognosis prediction. Lung cancer stem cell markers include surface markers and intracellular markers, and are associated with the stem potential and uncontrollable proliferation of tumor cells.

Cyclophilin A/CD147 Interaction: A Promising Target for Anticancer Therapy

Jang Mi Han et al.

Summary: The interaction between Cyclophilin A and CD147 plays a crucial role in multiple diseases, particularly in the progression of cancer. Their overexpression is associated with cancer cell growth, metastasis, therapeutic resistance, and stem cell-like properties, leading to poor prognosis in cancer patients. Therefore, targeting the Cyclophilin A/CD147 interaction may improve treatment outcomes for cancer patients.

Identification of Cyclophilin A as a Potential Anticancer Target of Novel Nargenicin A1 Analog in AGS Gastric Cancer Cells

Jang Mi Han et al.

Summary: The novel nargenicin A1 analog, 23-demethyl 8,13-deoxynargenicin, exhibits potential anti-cancer and anti-angiogenic activities against human gastric adenocarcinoma cells. Compound 9 targets cyclophilin A (CypA) and downregulates CD147-mediated MAPK signaling pathway, resulting in suppression of gastric cancer cell proliferation, migration, invasion, and angiogenesis. The interaction between compound 9 and CypA was confirmed through various assays, providing a new molecular basis for inhibiting gastric cancer progression.
