MMP-10 from M1 macrophages promotes pulmonary vascular remodeling and pulmonary arterial hypertension

Pei-Ling Chi et al.

Summary: This study found that levels of MMP-1 and MMP-10 were increased in the vessel wall, serum, and M1-polarized macrophages of PAH patients and PAH rodent models. MMP-10 promotes the malignant phenotype of PASMCs by promoting their proliferation and migration. Inhibition of STAT1 can suppress hypoxia-induced MMP-10 expression in M1-polarized macrophages.

Airway Macrophages Encompass Transcriptionally and Functionally Distinct Subsets Altered by Smoking

Maude Liegeois et al.

Summary: This study explores the heterogeneity of human alveolar macrophages (AMs) isolated from different groups, finding distinct subpopulations of AMs in BAL fluid based on autofluorescence levels. Single-cell RNA sequencing analyses reveal transcriptionally distinct clusters of classical and monocyte-derived AM enriched in smokers with and without COPD. Signs of gene signatures related to detoxification, oxidative stress, and proinflammatory responses are observed in smoking-associated clusters, indicating potential implications for COPD initiation or progression.

There and back again: Intracellular trafficking, release and recycling of matrix metalloproteinases

Sven Hey et al.

Summary: Matrix metalloproteinases play important roles in various physiological and pathological processes by cleaving different substrates, with their activities tightly regulated spatiotemporally. Intracellular trafficking, secretion, surface exposure, and recycling of MMPs involve complex molecular mechanisms and microtubule-dependent motility. Open questions in MMP motility may be significant in the future.

Summary: Pulmonary hypertension (PH) is a common complication of chronic obstructive pulmonary disease (COPD), but there are no blood biomarkers available to identify PH in COPD patients. This study investigated whether circulating angiogenic factors and cytokines could serve as biomarkers for COPD-PH patients. The results showed that elevated TIMP-1 may serve as a circulating biomarker for diagnosing PH among COPD patients.

Revisiting matrix metalloproteinase 12: its role in pathophysiology of asthma and related pulmonary diseases

Khalid Abd-Elaziz et al.

Summary: Matrix metalloproteinases (MMPs) are a family of zinc-dependent proteases with various biological and pathological activities. Increased levels of MMP-12 have been associated with inflammation and structural changes in the lungs in conditions such as asthma, COPD, and pulmonary fibrosis. Targeting MMP-12 may provide further insights into disease mechanisms and potential therapeutic strategies, but further research is needed to fully understand its role in these pulmonary diseases.

Multifunctional intracellular matrix metalloproteinases: implications in disease

Wesam Bassiouni et al.

Summary: Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that were initially discovered as proteases targeting extracellular proteins. However, recent research has revealed their crucial intracellular roles in various pathological conditions, including cardiovascular renal disorders, inflammation, and malignancy. In addition to MMP-2, at least eleven other MMPs have been identified to function intracellularly, highlighting the importance of understanding their roles in cellular processes and potential as therapeutic targets.

TIMP-3, an endogenous inhibitor of ADAMs and MMPs, shows potential advantages in therapeutic approaches. Through mass spectrometry analysis, this study reveals that high levels of TIMP-3 induce modifications in the cell surfaceome and identifies molecular pathways that can be deregulated through TIMP-3-based therapies.
