The Role of MicroRNAs in Pancreatitis Development and Progression

Hetvi R. R. Patel et al.

Summary: Pancreatitis is an inflammatory disease with high hospitalization and mortality rates. Recent studies have shown abnormal expression patterns of microRNAs in pancreatic tissues of patients with pancreatitis. MicroRNAs play a crucial role in regulating various aspects of pancreatic biology and have been implicated in the progression and development of the disease. They also interact with pancreatic stellate cells and immune cells, making them important in diagnosing and treating pancreatitis.

Probiotics counteract the expression of hepatic profibrotic genes via the attenuation of TGF-β/SMAD signaling and autophagy in hepatic stellate cells

Paulraj Kanmani et al.

Summary: This study found that LAB strains can attenuate liver fibrosis by modulating TGF-beta signaling. In vitro experiments showed that pretreatment with LAB strains reduced collagen deposition and upregulation of inflammatory markers in LX-2 cells stimulated by TGF-beta, and it also positively regulated autophagic and apoptotic associated proteins.

Disruption of pancreatic stellate cell myofibroblast phenotype promotes pancreatic tumor invasion

Elizabeth R. Murray et al.

Summary: PKN2 plays a critical role in the differentiation of pancreatic stellate cells into myofibroblast-like cancer-associated fibroblasts. Loss of PKN2 leads to reduced proliferation and contractility of pancreatic stellate cells, as well as a switch from myofibroblast to inflammatory cancer-associated fibroblast phenotype. Furthermore, deletion of PKN2 promotes invasive cancer cell outgrowth and induces more locally invasive pancreatic tumors. The absence of PKN2 in the pancreatic stroma predicts poor outcome in pancreatic and other solid human cancers.

Pirfenidone ameliorates chronic pancreatitis in mouse models through immune and cytokine modulation

Ejas Palathingal Bava et al.

Summary: The study aimed to evaluate the therapeutic efficacy of pirfenidone in mouse models of CP. The results suggest that pirfenidone improves CP-related changes in the pancreas, reduces fibrosis and inflammation. It also reduces the pro-fibrotic phenotype of macrophages and collagen secretion in pancreatic stellate cells.

Inflammatory-miR-301a circuitry drives mTOR and Stat3-dependent PSC activation in chronic pancreatitis and PanIN

Fugui Li et al.

Summary: miR-301a is highly expressed in activated pancreatic stellate cells (PSCs) and plays an important role in chronic pancreatitis and pancreatic tumor development. Downregulation of miR-301a can reduce pancreatic fibrosis and inhibit the proliferation and activation of PSCs. Aberrant expression of miR301a and Gadd45g restricts the interaction between PSCs and pancreatic cancer cells.

Miaomiao Li et al.

Summary: This study evaluated the therapeutic effect of the antioxidant, Mitoquinone (MitoQ), in the treatment of chronic pancreatitis (CP). The results demonstrated that MitoQ alleviated pancreatic fibrosis and inhibited oxidative stress in a CP mouse model. In addition, MitoQ inhibited the activation of pancreatic stellate cells and the development of fibrotic phenotypes in vitro.

Activation of pancreatic stellate cells attenuates intracellular Ca2+ signals due to downregulation of TRPA1 and protects against cell death induced by alcohol metabolites

Agnieszka A. Kusiak et al.

Summary: Alcohol abuse is a growing problem in developed societies and a leading cause of acute and chronic pancreatitis. Research has shown that quiescent pancreatic stellate cells are more sensitive to ethanol metabolites, while activated stellate cells can withstand noxious signals and deposit extracellular matrix components despite ongoing inflammation.

Targeting pancreatic stellate cells in chronic pancreatitis: Focus on therapeutic drugs and natural compounds

Yang Wu et al.

Summary: Chronic pancreatitis (CP) is a precancerous illness associated with pancreatic ductal adenocarcinoma (PDAC), but the mechanism of evolution is uncertain. CP is characterized by severe fibrosis caused by the activation of pancreatic stellate cells (PSCs). The current clinical therapeutic protocol lacks specific medicines for preventing and suppressing inflammation and fibrosis in CP. Research on targeting PSCs could lead to the development of novel therapies for pancreatic fibrosis.

Baicalin Ameliorates Pancreatic Fibrosis by Inhibiting the Activation of Pancreatic Stellate Cells in Mice with Chronic Pancreatitis

Jianwei Fan et al.

Summary: Baicalin has been shown to inhibit the activation of pancreatic stellate cells (PSCs), reduce pancreatic inflammation and fibrosis, and decrease the levels of various markers associated with fibrosis in the pancreas. It inhibits the TGF-beta 1/TGF-beta R1/TAK1/NF-kappa B signaling pathway, leading to decreased MCP-1 secretion and reduced infiltration of macrophages and inflammation cells in the pancreas. This study provides a basis for the potential use of baicalin in the prevention and treatment of chronic pancreatitis.
