CXCL9 Links Skin Inflammation and Fibrosis through CXCR3-Dependent Upregulation of Col1a1 in Fibroblasts

Jillian M. Richmond et al.

Summary: Morphea is a poorly studied disease characterized by initial inflammation followed by fibrosis of the skin and soft tissue. In this study, a mouse model was used to mimic the clinical and histological features of human morphea, and it was found that the CXCR3 chemokine axis plays a crucial role in the pathogenesis of cutaneous fibrosis. Fibroblasts were identified as the main source of CXCR3 ligands, and the fibrosis was shown to be dependent on CXCL9 and CXCR3. Furthermore, recombinant CXCL9 was found to contribute to fibrosis by inducing Col1a1 mRNA expression in mouse fibroblasts.

Gene Expression Signatures in Inflammatory and Sclerotic Morphea Skin and Sera Distinguish from Sclerosis

Henry W. Chen et al.

Summary: Morphea is a skin-directed process characterized by T helper 1 immune-mediated dysregulation, which contrasts with fibrotic signatures and systemic transcriptional changes associated with systemic sclerosis. The similarity between morphea and the inflammatory subset of systemic sclerosis on transcriptional profiling indicates that therapies under development for this subset of systemic sclerosis are also promising for treatment of morphea.

Single-Cell RNA Sequencing of Human Pluripotent Stem Cell-Derived Macrophages for Quality Control of The Cell Therapy Product

Hye-Yeong Jo et al.

Summary: Macrophages play important roles in maintaining tissue homeostasis, immune response, and tissue repair. They have potential therapeutic applications in treating inflammatory disorders and cancer. Human pluripotent stem cell (hPSC)-derived macrophages are considered as an alternative source of primary macrophages for clinical use. However, the quality of these hPSC-derived macrophages is still unclear. In this study, we evaluated the biological homogeneity of hPSC-derived macrophages and demonstrated the usefulness of single cell RNA sequencing data in quality control of cell-based therapy products.

LGR5 expressing skin fibroblasts define a major cellular hub perturbed in scleroderma

Chamutal Gur et al.

Summary: This study conducted single-cell genomic analysis of skin and blood samples from patients with systemic sclerosis (SSc) and healthy controls. The results showed immune compartment dysfunction in a specific subtype of diffuse SSc patients, and global dysregulation of the stromal compartment, particularly in a previously undefined subset of LGR5(+)-scleroderma-associated fibroblasts (ScAFs). The findings provide insights into disease development and may contribute to the development of biomarkers and therapeutic strategies.

Summary: Differential expression analysis in single-cell transcriptomics allows scientists to study cell-type-specific responses to biological perturbations. SFRP4 Expression Is Linked to Immune-Driven Fibrotic Conditions, Correlates with Skin and Lung Fibrosis in SSc and a Potential EMT Biomarker

Summary: SFRP4 expression is increased in skin fibrosis in both systemic sclerosis and sclerotic chronic graft versus host disease. In vitro studies suggest that SFRP4 may play a role in epithelial-mesenchymal transition. Serum levels of SFRP4 correlate with severity of skin and lung fibrosis in systemic sclerosis, suggesting it may serve as a biomarker of EMT.

Transcriptomic Evaluation of Juvenile Localized Scleroderma Skin With Histologic and Clinical Correlation

Christina Schutt et al.

Summary: Juvenile localized scleroderma (LS) is characterized by distinct genetic signatures among affected children, with correlations between skin inflammation scores and expression levels of certain genes. The study identified three different immunophenotype clusters in children with juvenile LS, providing insights into the underlying molecular pathways associated with the disease. Up-regulation of HLA class II genes in an inflammation-related cluster suggests a potential genetic basis for the predominant inflammatory features seen in these patients.
