TRPM1 promotes tumor progression in acral melanoma by activating the Ca2+/CaMKIId/AKT pathway

Chi-Che Hsieh et al.

Summary: TRPM1 is associated with tumor progression and malignancy in acral melanoma. Activation of the Ca2+/CaMKII8/AKT pathway by TRPM1 promotes melanoma progression, and inhibition of CaMKII may be a potential therapeutic strategy for acral melanomas with high expression of TRPM1.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Antagonism of the transient receptor potential melastatin-2 channel leads to targeted antitumor effects in primary human malignant melanoma cells

Shelby G. McKamey et al.

Summary: This study found that antagonism of the TRPM2 channel has antitumor effects in human melanoma cells, including those that are unresponsive to current treatments due to the expression of drug resistance genes. TRPM2 has a unique role in melanoma cells.

The human ion channel TRPM2 modulates cell survival in neuroblastoma through E2F1 and FOXM1

Iwona Hirschler-Laszkiewicz et al.

Summary: TRPM2 plays an important role in cell survival, mitochondrial function, and antioxidant response. Its deletion leads to decreased viability, cell cycle arrest, and increased DNA damage. Restoration of TRPM2 expression reduces tumor proliferation and increases sensitivity to doxorubicin through modulation of FOXM1, E2F1, and cell cycle/DNA repair proteins.

Summary: Transient receptor potential (TRP) proteins are ion channels that play a crucial role in controlling physiological functions and are potential therapeutic targets. Studying the role of TRP channels and utilizing new technologies can provide valuable insights for disease treatment. Fluorescent tracer combined with ion channel reader has been proven effective in analyzing ion channel activity, offering potential for drug discovery.

MMP9: A Tough Target for Targeted Therapy for Cancer

Katarzyna Augoff et al.

Summary: MMP9, a protease with diverse functions, plays a crucial role in tumorigenesis by regulating cancer cell migration, epithelial-to-mesenchymal transition, immune response, angiogenesis, and tumor microenvironment formation. However, the complicated regulation mechanism and high homology with other MMP family members make it difficult to develop effective and safe MMP9 inhibitors as anticancer drugs. The recent development of blocking antibodies shows promise, but further research is needed to fully understand the mechanisms of MMP9 in tumorigenesis and develop new therapies targeting MMP9.

TRPM2 Oxidation Activates Two Distinct Potassium Channels in Melanoma Cells through Intracellular Calcium Increase

Loretta Ferrera et al.

Summary: The response to oxidative stimulation in melanoma cells involves the activation of potassium currents and an increase in intracellular calcium levels, mediated by BK, KCa3.1, and TRPM2 channels. Differences in the expression levels of these channels in different cell lines may play a crucial role in melanoma progression. Targeting these ion channels pharmacologically could be a novel strategy to reduce the metastatic potential of melanoma cells alongside traditional treatments.
