Engineered disulfide reveals structural dynamics of locked SARS-CoV-2 spike

Summary: The spike protein of SARS-CoV-2 has different conformations, and the function of the locked conformation is unclear. In this study, a construct was engineered to stabilize the locked conformation of the spike protein. Two distinct locked-1 and locked-2 conformations were identified in the original SARS-CoV-2 strain, while only the locked-2 conformation remained after the D614G mutation. The locked conformations prevent premature opening of the receptor binding domain during virus assembly and release. This study provides a tool for further research on the locked conformations and their effect on S assembly and receptor binding domain dynamics.
