HPV-associated oropharyngeal cancer: epidemiology, molecular biology and clinical management

Matt Lechner et al.

Summary: The incidence of HPV-positive oropharyngeal squamous cell carcinoma (OPSCC) is rapidly increasing, leading to ongoing research in treatment de-escalation and emerging therapies. There is a need to identify and validate diagnostic biomarkers for early detection of this disease.

Promising Biomarkers in Head and Neck Cancer: The Most Clinically Important miRNAs

Arsinoe C. Thomaidou et al.

Summary: Head and neck cancers comprise a heterogeneous group of tumors with diverse etiologic factors. Certain miRNAs have been identified as potential biomarkers for the diagnosis and prognosis of HNC. Ongoing clinical trials aim to determine the utility of miRNAs in HNC.

Impact of Alcohol and Smoking on Outcomes of HPV-Related Oropharyngeal Cancer

Yu-Hsuan Lai et al.

Summary: This study evaluated the impact of adverse lifestyle factors on outcomes in patients with HPV-related oropharyngeal squamous cell carcinoma (OPSCC). Alcohol use was found to increase the risk of disease relapse in HPV-positive OPSCC patients, while heavy smoking age >= 30 pack-years was associated with increased risk of death and disease relapse in OPSCC patients. The combination of alcohol use and heavy smoking further increased the risk of disease relapse in HPV-positive OPSCC patients. These findings suggest that adverse lifestyle factors may influence the treatment outcomes of HPV-related OPSCC.

Prognostic impact of additional HPV diagnostics in 102 patients with p16-stratified advanced oropharyngeal squamous cell carcinoma

Bernhard G. Weiss et al.

Summary: The study found that p16 and HPV status may not be equivalent markers for a better prognosis, and may not significantly affect the survival rate of patients. Differences were observed in survival rates between smokers and non-smokers during treatment.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Distinguishable Prognostic miRNA Signatures of Head and Neck Squamous Cell Cancer With or Without HPV Infection

Xiao-jie Luo et al.

Summary: The study developed miRNA signatures for both HPV+ and HPV- HNSCC to predict patients' prognosis, and explored their potential biological functions. Combined results suggested that HPV+ HNSCC patients with low signature scores had better immunity and treatment sensitivity, while HPV- HNSCC patients with high scores had poor prognosis due to treatment resistance and cell metabolism dysregulation. Identified signatures for HPV+ and HPV- HNSCC are significant for patient outcomes and may serve as new biomarkers and therapeutic targets.
