Structural basis of efficacy-driven ligand selectivity at GPCRs

Serendipitous discovery of ligands that stimulate target receptors more strongly than closely related receptors provides a solution. This study reveals the structural basis for the efficacy-driven selectivity of xanomeline, a clinical drug candidate, between closely related muscarinic acetylcholine receptors (mAChRs), using atomic-level simulations. The results suggest strategies for rational design of ligands achieving efficacy-driven selectivity for G-protein-coupled receptors.

Molecular mechanism of biased signaling at the kappa opioid receptor

Effect of Ligands and Transducers on the Neurotensin Receptor 1 Conformational Ensemble

Muscarinic acetylcholine receptors for psychotic disorders: bench-side to clinic

Summary: Modern interest in muscarinic acetylcholine receptor (mAChR) activators for schizophrenia began in the 1990s when xanomeline, an M1/M4-preferring mAChR agonist developed for cognitive symptoms of Alzheimer's disease (AD), had unexpected antipsychotic activity. However, strategies to address tolerability concerns associated with activation of peripheral mAChRs were not available at that time. The discovery of specific targeted ligands and combination treatments to reduce peripheral mAChR engagement have advanced the potential of mAChR activators as effective treatments for psychotic disorders.

Structures in G proteins important for subtype selective receptor binding and subsequent activation

These determinants differ in their impact on selective binding to GPCRs and subsequent activation depending on the specific receptor, providing new insights into the molecular basis of G protein-coupling selectivity even beyond the G alpha C-terminus. Jelinek, Mosslein & Bunemann conclude that various regions, especially outside of the C-terminal tail of the G alpha subunit, collectively affect G-protein-coupling selectivity.
