Pharmacological Modulation of the MIP-1 Family and Their Receptors Reduces Neuropathic Pain Symptoms and Influences Morphine Analgesia: Evidence from a Mouse Model

Agata Ciechanowska et al.

Summary: The study examined the effects of pharmacological modulation of chemokines and their receptors on hypersensitivity after nerve injury. The results showed increased levels of CCL3 and CCL9 after nerve injury, and treatment with neutralizing antibodies or antagonists for these chemokines relieved neuropathic pain symptoms and enhanced morphine analgesia. These findings suggest that CCL3 and CCL9 play important roles in neuropathic pain and opioid analgesia, and CCR1 and CCR5 may serve as potential targets for neuropathy treatment.

New insights into the analgesic properties of the XCL1/XCR1 and XCL1/ITGA9 axes modulation under neuropathic pain conditions-evidence from animal studies

Agata Ciechanowska et al.

Summary: This study investigates the involvement of chemokine-C-motif ligand 1 (XCL1) and its receptors, XCR1 and ITGA9, in nociceptive transmission. The results show that the blockade/neutralization of both XCR1 and ITGA9 receptors can reverse hypersensitivity and improve opioid analgesia. The findings suggest that blocking the XCL1-ITGA9 interaction may serve as a novel strategy for the treatment of neuropathic pain in combination with opioids.

Astaxanthin alleviates neuropathic pain by inhibiting the MAPKs and NF-κB pathways

Lin Zhao et al.

Summary: Neuropathic pain is a complex condition that can last a lifetime and has a significant negative impact on quality of life. Astaxanthin, a natural marine compound, has shown promise in reducing spinal nerve ligation (SNL) damage by inhibiting inflammatory responses and signaling pathways, suggesting its potential as a candidate for pain management in neuropathic pain.

Intrathecal injection of ozone alleviates CCI-induced neuropathic pain via the GluR6-NF-κB/p65 signalling pathway in rats

Weiguang Zhang et al.

Summary: The study revealed that ozone may alleviate neuropathic pain in rats with chronic constriction injury of the sciatic nerve by modulating the spinal GluR6-NF-kappa B/p65 signaling pathway. Additionally, ozone was found to inhibit pain behaviors and suppress the expression of spinal inflammatory mediators, indicating its potential as a therapeutic intervention for neuropathic pain.

Summary: The recent findings have highlighted the roles of the CXC chemokine family in the mechanisms of neuropathic pain. The studies provide evidence that antagonists of CXCR2 and CXCR3 can effectively attenuate mechanical and thermal hypersensitivity in rats after nerve injury. Additionally, the antagonists showed analgesic effects on reducing levels of pronociceptive chemokines and interleukins in the spinal cord and dorsal root ganglia.
