Properties of FDA-approved small molecule protein kinase inhibitors: A 2023 update

Summary: Due to protein kinase dysregulation, it has become crucial to target this enzyme family in many diseases, especially cancer. There are 72 FDA-approved drugs that target different protein kinases, with three of them receiving approval in 2022. These drugs target various types of protein kinases, and the majority are used to treat neoplasms and inflammatory diseases. This review provides a summary of the physicochemical properties of these drugs, including lipophilic efficiency and ligand efficiency.

Chlorantraniliprole in foods: Determination, dissipation and decontamination

Summary: Chlorantraniliprole (CAP) is the first widely used anthranilic diamide insecticide that targets ryanodine receptors, but its excessive use can result in persistent contamination on treated foods and negative effects on human health. This review focuses on the analysis of CAP residues in foods using chromatographic methods, with QuEChERS being the most commonly used sample preparation strategy and liquid chromatography tandem mass spectrometry as the predominant analytical method. Furthermore, the dissipation pattern of CAP on foods is summarized, revealing its fast dissipation on plants in open-field environments. Common culinary cleaning methods are effective in removing CAP from vegetables. Finally, suggestions are made for further advancements.

Summary: This study investigated molecular thermodynamic properties of ophthalmic drugs to establish a framework for product development. By analyzing the distribution of molecular properties of 145 ophthalmic drugs, key molecular parameters important for ocular permeability and absorption were identified. These parameter limits are proposed as rules of thumb for assessing developmental risks of topical ophthalmic drugs.

Fabrication of Isopropanolamine-Decorated Coumarin Derivatives as Novel Quorum Sensing Inhibitors to Suppress Plant Bacterial Disease

Summary: This study designed and synthesized a series of compounds that interfere with bacterial quorum sensing (QS), and identified a compound A5 with antibacterial activity. Compound A5 was found to inhibit biofilm formation, suppress bacterial virulence factors, and decrease the expression of QS-related genes. It also showed acceptable control effect against Xanthomonas oryzaepv.oryzae (Xoo) in vivo.

Summary: In this study, a series of natural pterostilbene derivatives were designed and assessed for their antibacterial potency and antibiofilm ability. Compound C-1 exhibited excellent antimicrobial activity and biofilm inhibition against Xanthomonas oryzae pv. oryzae. C-1 also reduced biofilm formation, inhibited bacterial pathogenicity and other virulence factors.

Compound 10h showed strong inhibitory activity against Gibberella zeae and control activity against corn scab. It could also inhibit the enzymatic activity of SDH in the G. z. strain.

Dysregulation of ClpP by Small-Molecule Activators Used Against Xanthomonas oryzae pv. oryzae Infections

Summary: By determining the crystal structures of XooClpP and its mutant bound with ADEP4, this work revealed the action modes of XooClpP assemblies and XooFtsZ degradation in the presence of small-molecule activators such as ONC212 and ADEP4. An antibacterial assessment showed that ONC212 has excellent activity against Xoo and can prevent rice bacterial leaf blight in vivo. These unique antibacterial effects of small-molecule activators of XooClpP represent a potential strategy for the development of agricultural antibiotics targeting bacterial ClpP.

Synthesis and Antifungal Activity of Pyrimidine Derivatives Containing an Amide Moiety
