Association of CSF, Plasma, and Imaging Markers of Neurodegeneration With Clinical Progression in People With Subjective Cognitive Decline

Jarith L. Ebenau et al.

Summary: This study investigated the association of multiple neurodegeneration biomarkers with clinical progression and cognitive decline in individuals with subjective cognitive decline. The results showed that these biomarkers individually predicted clinical progression and cognitive decline, with hippocampal volume, serum neurofilament light, and serum glial fibrillary acidic protein showing the highest predictive value.

Altered central and blood glutathione in Alzheimer's disease and mild cognitive impairment: a meta-analysis

Jinghan Jenny Chen et al.

Summary: This meta-analysis examined the levels of GSH in the brain and blood of patients with AD and MCI compared to healthy controls. The results showed that brain GSH did not differ significantly in AD and MCI compared to healthy controls. However, lower brain GSH was observed in AD and MCI patients in studies using MEGA-PRESS. In addition, AD patients had lower levels of intracellular and extracellular blood GSH, and MCI patients had lower levels of intracellular blood GSH.

Dipeptide of ψ-GSH Inhibits Oxidative Stress and Neuroinflammation in an Alzheimer's Disease Mouse Model

Abbas Raza et al.

Summary: Supplementation of gamma-GT-resistant GSH analogue psi-GSH and its precursor dipeptide 2 showed potential as a therapeutic for Alzheimer's disease. Compound 2 exhibited antioxidant properties and protection against A beta(1-42)-aggregates, resulting in cognitive benefits and mitigation of oxidative stress and inflammatory pathology in an acute AD model.

Pharmacological intervention in a transgenic mouse model improves Alzheimer's-associated pathological phenotype: Involvement of proteasome activation

Aleksandra N. Mladenovic Djordjevic et al.

Summary: This study demonstrated the potential therapeutic effect of proteasome activation in alleviating Alzheimer's disease pathology in a transgenic mouse model. Activation of proteasome led to reduced amyloid-beta load, improved motor functions, reduced anxiety, and reduced frailty level, opening up new directions for future therapeutic interventions targeting proteasome-mediated proteolysis enhancement.

Quercetin stimulates the non-amyloidogenic pathway via activation of ADAM10 and ADAM17 gene expression in aluminum chloride-induced Alzheimer's disease rat model

Asmaa M. Elfiky et al.

Summary: The present study demonstrated that quercetin has neuroprotective effects on Alzheimer's disease, attenuating cognitive impairment, neurotransmitter dysfunction and amyloid beta aggregation in the hippocampus through modulation of gene expression. Activation of ADAM10 and ADAM17 by quercetin may serve as potential therapeutic targets for counteracting Alzheimer's disease progression.

Glutathione in the Brain

Koji Aoyama

Summary: Glutathione (GSH) depletion in neurons is common in neurodegenerative diseases and can lead to neurodegeneration. Excitatory amino acid carrier 1 (EAAC1) plays a central role in regulating neuronal GSH production, making it a potential target for therapeutic strategies in these diseases.

Secreted Amyloid Precursor Protein-Alpha Enhances LTP Through the Synthesis and Trafficking of Ca2+-Permeable AMPA Receptors

Rhys W. Livingstone et al.

Summary: sAPP alpha enhances hippocampal LTP by promoting the cell-surface expression of GluA1 AMPARs and reducing GluA2 levels, affecting the localization and internalization of AMPARs at synapses, ultimately increasing synaptic levels of GluA1/2 AMPARs.
