Summary: This study found that MIR630 represses the expression of NANOG through transcriptional and post-transcriptional regulation, leading to the differentiation of NT2/D1 cells. MIR630 may have multiple binding sites in the 3'UTR of NANOG and may act together with other miRNAs to maximally suppress NANOG expression.

Reciprocal epigenetic remodeling controls testicular cancer hypersensitivity to hypomethylating agents and chemotherapy

Ratnakar Singh et al.

Summary: TGCTs exhibit reciprocal sensitivity to hypomethylating agents and cisplatin, with resistant cells showing opposite responses. The downregulation of polycomb target gene expression in hypomethylating agent resistant cells contrasts with the upregulation in cisplatin resistant cells, indicating a complex relationship between epigenetic changes and drug sensitivity in TGCTs.

The component of the m6A writer complex VIRMA is implicated in aggressive tumor phenotype, DNA damage response and cisplatin resistance in germ cell tumors

Vera Miranda-Goncalves et al.

Summary: The study found significant differences in the expression of various m(6)A modifiers in cell lines representing different classes of GCTs, as well as changes after treatment with all-trans retinoic acid. Knockdown of VIRMA resulted in disruption of the methyltransferase complex, decreased m(6)A abundance, reduced tumor aggressiveness, and increased sensitivity to cisplatin treatment both in vitro and in vivo. Enhanced response to cisplatin after VIRMA knockdown was associated with increased DNA damage and downregulation of specific genes related to DNA repair.
