Prognostic role of inflammatory cytokines and novel adipokines in acute myocardial infarction: An updated and comprehensive review

Andreas Mitsis et al.

Summary: Acute myocardial infarction (AMI) is a significant global cause of morbidity and mortality. Inflammation response plays a key role in the peri-AMI period, affecting ischaemia-reperfusion injury, cardiac remodelling, infarct size, and prognosis. This article provides an updated overview of cytokines and adipokines involved in AMI pathophysiology, summarizing their prognostic role. Elevated levels of proinflammatory cytokines and adipokines are associated with high mortality and morbidity, while increased anti-inflammatory cytokines and adipokines may have a better prognosis. Incorporating these inflammation biomarkers into prognostic models can improve the management of AMI patients.

Targeting the CCL2-CCR2 axis for atheroprotection

Marios K. Georgakis et al.

Summary: Decades of research have established atherosclerosis as an inflammatory disease. Clinical trials have shown the efficacy of anti-inflammatory strategies in reducing cardiovascular events, but there is a need for new anti-inflammatory agents that target atherosclerosis-specific immune mechanisms. The CCL2-CCR2 axis has therapeutic potential in human atherosclerosis. However, there are challenges and opportunities related to pharmacological targeting of this pathway that need further investigation and discussion.

Realigning the LIGHT signaling network to control dysregulated inflammation

Carl F. F. Ware et al.

Summary: Advances in understanding the TNFSF and its role in immune responses have shed light on the pathogenesis of infectious and autoimmune diseases. LIGHT, as a key modulator of innate and adaptive immunity, forms an immune regulatory network with other proteins. Dysregulation of this network has been implicated in diseases such as COVID-19 pneumonia and inflammatory bowel diseases, suggesting the potential of target-modifying therapeutics in disease intervention.

Summary: Cardiovascular pathologies are leading causes of mortality worldwide, with atherosclerosis being a chronic disease associated with serious complications. Oxidized LDL may have both anti-inflammatory and pro-inflammatory properties, triggering inflammation through macrophage activation and other cells.

TNF leads to mtDNA release and cGAS/STING-dependent interferon responses that support inflammatory arthritis

Joschka Willemsen et al.

Summary: The study elucidates a molecular mechanism linking TNF to type-I interferon signaling, demonstrating that TNF triggers a cGAS-STING-dependent interferon response and inhibits PINK1-mediated mitophagy. This leads to altered mitochondrial function, increased cytosolic mtDNA levels, and a transcriptional response that mimics macrophages from rheumatoid arthritis patients. Additionally, in an inflammatory arthritis mouse model, cGAS deficiency blocked interferon responses and reduced inflammatory cell infiltration and joint swelling. Therapeutic targeting of cGAS/STING in TNF-driven diseases may offer potential benefits.

The stressosome, a caspase-8-activating signalling complex assembled in response to cell stress in an ATG5-mediated manner

Katarzyna Mnich et al.

Summary: Stress-induced apoptosis is primarily mediated through the intrinsic pathway involving caspase-9. A protein complex termed the 'stressosome' containing ATG5-ATG12, FADD, and caspase-8 was found to be activated by various stress stimuli in caspase-9-deficient cells. However, only ER stress and heat shock led to stressosome-dependent cell death. In silico modeling proposed the structure of the stressosome complex, with FADD acting as an adaptor protein interacting with pro-caspase-8 and ATG5-ATG12.
