Insights into the pathogenesis of hereditary angioedema using genetic sequencing and recombinant protein expression analyses

Zhen Ren et al.

Summary: This study investigated the influence of SERPING1 variants on C1 esterase inhibitor (C1-INH) expression, structure, and function in 20 patients with hereditary angioedema (HAE). Through whole-exome sequencing and whole-genome sequencing, 11 heterozygous variants were identified, including 5 pathogenic variants and 6 variants of uncertain significance. The study revealed that certain variants affect the synthesis, folding, and function of C1-INH, and that the compound variant P399A/L349F is associated with a more severe clinical phenotype.

Therapeutic application of Sertoli cells for treatment of various diseases

Rachel L. Washburn et al.

Summary: Sertoli cells, as immune privileged cells in the testis, provide immune protection to maturing germ cells. They have potential therapeutic applications in creating tolerogenic environments, protecting co-grafted cells, and delivering therapeutic proteins through gene therapy. Studies have shown their potential use in treating diabetes, neurodegenerative disorders, and restoration of spermatogenesis.

Influence of Von Willebrand Disease (VWD) and pregnancy on the expression of angiogenic factors in the porcine female reproductive tract

Rabea Moeller et al.

Summary: Von Willebrand Disease (VWD) is a heritable disorder caused by defects of the Von Willebrand Factor (VWF). In a porcine model, it was found that CCN2 gene expression was upregulated in the uterus and ovary of non-pregnant VWD-affected pigs, and F8 gene expression was significantly reduced in these organs. During pregnancy, CCN2 expression increased in wildtype pig uteri but hardly changed in those of pregnant pigs affected by VWD.

Mouse Sertoli Cells Inhibit Humoral-Based Immunity

Rachel L. Washburn et al.

Summary: Transplantation is a common treatment for various diseases, but the immune system often rejects the graft unless immunosuppressants are used. Sertoli cells (SCs), a type of testicular cells, can survive long-term after transplantation without immunosuppression. This study aims to understand how SCs regulate the immune response. The results suggest that SCs inhibit complement-mediated killing, which is an important mechanism in their immune regulation.

Complement Receptors and Their Role in Leukocyte Recruitment and Phagocytosis

Sofie Vandendriessche et al.

Summary: The complement system plays a crucial role in controlling microorganisms and removing dead cells, while also stimulating adaptive immune responses and tissue regeneration. Complement receptors on leukocytes and parenchymal cells are essential for recognizing complement-derived molecules and promoting immune responses.

New aspects in the regulation of human B cell functions by complement receptors CR1, CR2, CR3 and CR4

Anna Erdei et al.

Summary: Complement is known to regulate antibody responses and additional B cell functions such as cytokine production and antigen presentation. These activities are mediated by receptors interacting with activation fragments of C3, attached to antigens and immune complexes. This review focuses on human B lymphocytes and highlights important differences between human and mouse systems.

Complement Factor C5a Inhibits Apoptosis of Neutrophils-A Mechanism in Polytrauma?

Christian Ehrnthaller et al.

Summary: Life-threatening polytrauma results in early activation of the complement and apoptotic system in PMN, with polytrauma patients showing reduced apoptotic rates up to 10 days post trauma compared to healthy controls. Specific blockade of the C5a receptor 1 (C5aR1) on PMN reversed C5a-induced resistance to apoptosis, suggesting a potential therapeutic role of the C5a/C5aR1 interaction on PMN apoptosis after polytrauma.
