FAM19A4/miR124-2 Methylation Testing and Human Papillomavirus (HPV) 16/18 Genotyping in HPV-Positive Women Under the Age of 30 Years

Frederique J. Vink et al.

Summary: The FAM19A4/miR124-2 methylation test can detect nonproductive, transforming CIN2/3 lesions with high specificity in HPV-positive women under 30 years old, providing valuable information for treatment decisions.

Clinical Regression of High-Grade Cervical Intraepithelial Neoplasia Is Associated With Absence of FAM19A4/miR124-2 DNA Methylation (CONCERVE Study)

Wieke W. Kremer et al.

Summary: This study evaluated the prognostic value of FAM19A4/miR124-2 methylation in untreated CIN2/3 patients for clinical regression. The results showed that most untreated CIN2/3 patients with negative baseline FAM19A4/miR124-2 methylation showed clinical regression, indicating that methylation, combined with cytology or HPV genotyping, can support a wait-and-see policy for CIN2/3 patients.

Detection of Host Cell Gene/HPV DNA Methylation Markers: A Promising Triage Approach for Cervical Cancer

Lingyi Zhang et al.

Summary: Cervical cancer is common in low- and middle-income countries. Early screening plays a crucial role in prevention and treatment. Detection of host cell gene and/or HPV DNA methylation shows promise in early triage of cervical cancer. However, suitable genes or combinations for large-scale screening and triage still need to be determined.

Comparison of Expectant and Excisional/Ablative Management of Cervical Intraepithelial Neoplasia Grade 2 (CIN2) in the Era of HPV Testing

Summary: This study aimed to investigate the outcomes of conservative and excisional/ablative treatment for cervical intraepithelial neoplasia grade 2 (CIN2) after introducing virological test of cure. The results showed that conservative management is a reasonable and effective strategy for appropriately selected women with CIN2, with high rates of histological and virological regression.

Comparative effectiveness and risk of preterm birth of local treatments for cervical intraepithelial neoplasia and stage IA1 cervical cancer: a systematic review and network meta-analysis

Antonios Athanasiou et al.

Summary: This systematic review and network meta-analysis aimed to determine the risks of treatment failure and preterm birth associated with various treatment techniques for cervical intraepithelial neoplasia (CIN). The findings suggest that more radical excisional techniques reduce the risk of treatment failure but increase the risk of subsequent preterm birth. Ablative treatments probably do not increase the risk of preterm birth but are associated with higher failure rates than excisional techniques.

Adjunctive use of p16 immunohistochemistry for optimizing management of CIN lesions in a high-risk human papillomavirus-positive population

Renee M. F. Ebisch et al.

Summary: This study shows that adjunctive use of p16 immunohistochemistry to hematoxylin and eosin (H&E) morphology reduces the number of CIN1 and CIN2 classifications with a proportional increase in no CIN and CIN3 diagnoses. The pathologists felt more confident in classifying the material with H&E and p16 immunohistochemistry than by using H&E alone, particularly during assessment of small biopsies.

Classification of high-grade cervical intraepithelial neoplasia by p16ink4a, Ki-67, HPV E4 and FAM19A4/miR124-2 methylation status demonstrates considerable heterogeneity with potential consequences for management

Frederique J. Vink et al.

Summary: This study evaluates the immunohistochemical expression of p16(ink4a) and Ki-67, known as the immunoscore (IS), in relation to HPV E4 expression and FAM19A4/miR124-2 methylation in high-grade CIN lesions. The findings suggest that these biomarker profiles may help clinicians in decision-making regarding immediate treatment or observation to avoid unnecessary overtreatment of high-grade CIN lesions.

Genetic and Epigenetic Variations of HPV52 in Cervical Precancer

Katharine J. Bee et al.

Summary: This study aimed to identify genetic and epigenetic changes in HPV type 52 associated with high-grade cervical precancer and cancer. Increased methylation at CpG site 5615 in HPV52 L1 region was found to be significantly associated with CIN3, while specific SNPs present in sublineage C2 were linked to a higher risk of CIN3. These findings improve the potential for early diagnosis of cervical neoplasia caused by HPV52.
