Lipid reprogramming induced by the TFEB-ERRα axis enhanced membrane fluidity to promote EC progression

Xiaodan Mao et al.

Summary: This study reveals that lipid reprogramming induced by the TFEB-ERRα axis enhances the progression of endometrial cancer through EMT signaling. The TFEB-ERRα axis increases membrane fluidity, promotes pseudopod formation, and enhances cell invasion. TFEB and ERRα are highly expressed in EC patients and correlated with EC progression. Furthermore, the TFEB-ERRα axis affects lipid metabolism by altering mitochondrial function.

Role of HIF-1α/ERRα in Enhancing Cancer Cell Metabolism and Promoting Resistance of Endometrial Cancer Cells to Pyroptosis

Pingping Su et al.

Summary: Oxygen is crucial for energy metabolism in tumors, which often occur in a hypoxic microenvironment. The interaction between HIF-1 alpha and ERR alpha plays a key role in regulating the metabolic and functional changes in cancer cells. This review gives an overview of the tumor metabolic reprogramming involving HIF-1 alpha/ERR alpha and explores their role in promoting tumor growth adaptation and pyroptosis resistance.

Mendelian randomization analyses suggest a role for cholesterol in the development of endometrial cancer

Pik-Fang Kho et al.

Summary: This study used Mendelian randomization analysis to investigate the relationship between blood lipids and endometrial cancer risk. Genetically elevated LDL cholesterol levels were associated with reduced risk of endometrial cancer, while higher predicted HDL cholesterol levels were linked to increased risk of non-endometrioid endometrial cancer. Obesity may influence these associations, but triglycerides showed no evidence of involvement in endometrial cancer development.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Cholesterol and Sphingolipid Enriched Lipid Rafts as Therapeutic Targets in Cancer

Michela Codini et al.

Summary: Lipid rafts are critical cell membrane platforms enriched in sphingolipids and cholesterol, playing roles in various cellular processes, including cancer. Recent findings on cholesterol, sphingolipids, and lipid rafts in cellular and nuclear membranes provide insights into using lipid rafts as biomolecular targets for developing innovative therapeutic strategies in cancer.

Summary: This study conducted a multi-omics analysis to investigate the expression and potential roles of matrix metalloproteinases in colorectal cancer. The findings suggest that MMPs could be targeted in colon cancer treatment and serve as novel biomarkers, especially for their involvement in the immune response. Further research is needed to explore the detailed biological functions and molecular mechanisms of MMPs in CRC, considering their expression and regulation in different tissues.

Cholesterol-Lowering Drugs on Akt Signaling for Prevention of Tumorigenesis

Navneet Kumar et al.

Summary: Cholesterol accumulation in cancer cells is associated with tumor development, while cholesterol-lowering drugs have been found to play a role in the prevention and treatment of various cancers. Dysregulation of Akt in cancer is common and targeting Akt is considered an effective approach for cancer therapy. Cholesterol-lowering drugs affect Akt expression and activation in cancer cells, showing anticancer activity in different types of cancers.

Cholesterol-Induced Metabolic Reprogramming in Breast Cancer Cells Is Mediated via the ERRα Pathway

Faegheh Ghanbari et al.

Summary: This study demonstrates that obesity and high blood cholesterol levels are associated with an increased risk of recurrence and higher mortality in breast cancer patients by altering metabolic programming in breast cancer cells. Cholesterol acts as an endogenous agonist of ERR alpha, regulating cellular metabolism. The findings provide mechanistic insights into the link between cholesterol/obesity and metabolic reprogramming in breast cancer, potentially identifying therapeutic targets.
