Isolation and Characterization of Novel Lytic Bacteriophage vB_RsoP_BMB50 infecting Ralstonia solanacearum

Summary: In this study, a novel lytic bacteriophage named vB_RsoP_BMB50 was isolated from tomato fields in Dalian, China. It belonged to the subfamily Okabevirinae, Autographiviridae family, and order Caudovirales. The phage exhibited a large burst size, high temperature stability, and strong pH tolerance. Comparative analyses revealed that vB_RsoP_BMB50 represented a novel Ralstonia phage genus.

Complete Genome Sequence of a Novel Bacteriophage RpY1 Infecting Ralstonia solanacearum Strains

Summary: In this study, 72 phages were isolated from various crop cultivated soils in Korea, with phage RpY1 chosen for further investigation due to its specificity for the targeted host. RpY1 was identified as a member of Podoviridae with a head measuring 60-70 nm, a genome size of 43,284 bp, showing no homology to known phages except for DU_RP_II infecting R. solanacearum.

A rapid competitive method for bacteriophage genomic DNA extraction

Summary: A rapid and cost-effective method for phage DNA extraction was introduced in this study, taking less than 10 minutes to complete. The acetone precipitation and extraction by potassium iodide or sodium iodide were determined to be the best methods for phage DNA extraction based on time, cost, and quality of obtained DNA. The extracted genomic DNA using this method is suitable for phage genomic analysis such as restriction enzyme studies, DNA library preparation, and next-generation sequencing.
