Summary: Pancreatic ductal adenocarcinoma (PDAC) is a deadly cancer with poor response to current treatments. The protein Ref-1, which has redox signaling function, plays a role in PDAC signaling and has targets like RelA and STAT3. In this study, the researchers found that when RelA is deficient, PDAC cells are resistant to Ref-1 redox inhibitors, but sensitivity is restored when RelA is added back. IL-8, FOSB, and c-Jun expression were significantly decreased with Ref-1 inhibitors. PRDX1 also contributed to the cellular response to Ref-1 inhibitors, and the killing mechanism of Ref-1 inhibition was mediated through RelA, not STAT3.
