Conserved angio-immune subtypes of the tumor microenvironment predict response to immune checkpoint blockade therapy

Summary: Immune checkpoint blockade (ICB) therapy has revolutionized cancer treatment, but only a fraction of patients respond to it. This study identifies three distinct angio-immune tumor microenvironment (TME) subtypes conserved across multiple non-hematological cancers. Patients with TME showing low angiogenesis and strong anti-tumor immunity have the most significant responses to ICB therapy. The study suggests incorporating baseline angio-immune scores for future ICB treatment strategies.

CD36-mediated ferroptosis dampens intratumoral CD8+ T cell effector function and impairs their antitumor ability

Summary: Understanding the mechanisms of how T cells become dysfunctional in a tumor microenvironment is crucial for cancer immunotherapy. This study found that CD36 expression in tumor-infiltrating CD8(+) T cells, induced by TME cholesterol, is associated with tumor progression and poor survival, and that genetic ablation of Cd36 in these T cells leads to enhanced tumor eradication. Targeting CD36 or inhibiting ferroptosis could restore T cell function and enhance antitumor efficacy, especially in combination with anti-PD-1 antibodies.

Metabolic flexibility determines human NK cell functional fate in the tumor microenvironment

Summary: NK cells play a central role in anti-tumor immunity, but their dysfunction in the tumor microenvironment is a major barrier for cancer immunotherapies. Research showed that human NK cell dysfunction in the tumor microenvironment is caused by suppression of glucose metabolism, and activating the antioxidant pathway can restore NK cell function and enhance anti-tumor activity. Additionally, expanded NK cells with metabolic flexibility not only sustained metabolic fitness but also improved tumor killing in response to nutrient deprivation, showing the potential for exploiting metabolic flexibility in cancer immunotherapy.
