Progression of established non-diabetic chronic kidney disease is halted by metformin treatment in rats

Raphaelle Corremans et al.

Summary: This study evaluated the therapeutic efficacy of metformin and canagliflozin in slowing the progression of non-diabetic CKD. Metformin showed protective effects against kidney impairment, while canagliflozin did not. Proteomic analysis revealed that metformin's kidney-protective effect was associated with the activation of the Hippo signaling pathway.

The extract of Polygala fallax Hemsl. slows the progression of diabetic nephropathy by targeting TLR4 anti-inflammation and MMP-2/9-mediated anti-fibrosis in vitro

Minhui Wang et al.

Summary: This study aimed to investigate the effects and mechanisms of Polygala fallax extract (EPF) on high glucose-induced human glomerular mesangial cell (HMC) injury, inflammation, fibrosis, and apoptosis. The results showed that EPF could reverse the hyperproliferation and apoptosis of HMCs induced by high glucose, inhibit the increase in inflammatory factors, and downregulate the expression of TLR4/NF-xB pathway and its downstream factors.

YAP mediates the interaction between the Hippo and PI3K/Akt pathways in mesangial cell proliferation in diabetic nephropathy

Xuan Qian et al.

Summary: Under diabetic conditions, the Hippo pathway is inhibited, leading to YAP activation and promoting mesangial cell proliferation. The PI3K/Akt pathway is activated through the regulatory effect of YAP on its repressor, PTEN. Activation of the PI3K/Akt pathway inhibits the Hippo pathway, resulting in nuclear YAP accumulation and accelerating mesangial cell proliferation and DN formation.

Metformin regulates inflammation and fibrosis in diabetic kidney disease through TNC/TLR4/NF-κB/miR-155-5p inflammatory loop

Yang Zhou et al.

Summary: Type 2 diabetes mellitus (T2DM) is increasing globally, with its complications, particularly diabetic kidney disease (DKD), also on the rise. Increased levels of tenascin-C (TNC) are associated with UACR and hypertension in T2DM patients. In both a rat model of DKD and rat mesangial cells cultured under high glucose conditions, TNC expression levels were elevated. TNC was found to regulate miR-155-5p expression through the TLR4/NF-kappa B p65 pathway, affecting inflammation and fibrosis processes in DKD.

Metformin attenuates diabetic renal injury via the AMPK-autophagy axis

Tingli Sun et al.

Summary: This study found that metformin could reduce kidney damage caused by diabetic nephropathy (DN) by activating the AMPK-mTOR-autophagy axis, resulting in decreased oxidative damage, fibrosis, and inflammatory response. This suggests that metformin may be considered as a potential target in the treatment of DN.

The Effect of Metformin in Diabetic and Non-Diabetic Rats with Experimentally-Induced Chronic Kidney Disease

Mohammed Al Za'abi et al.

Summary: The study showed that metformin can effectively alleviate renal damage induced by adenine and STZ, especially when given together. This indicates that metformin may have an inhibitory effect on the progression of CKD in both diabetic and non-diabetic rats.

A novel polysaccharide obtained from Siraitia grosvenorii alleviates inflammatory responses in a diabetic nephropathy mouse model via the TLR4-NF-κB pathway

Pin Gong et al.

Summary: SGP-1-1 exhibited good inflammation-modulating activity in DN mouse models, alleviating cellular inflammation and tissue damage. Metabolomics analysis revealed potential biomarkers related to DN, and SGP-1-1 inhibited inflammation by regulating the TLR4-NF-kappa B pathway while promoting antioxidant activity.
