Cellular Plasticity, Reprogramming, and Regeneration: Metaplasia in the Stomach and Beyond

James R. Goldenring et al.

Summary: The mucosa of the body of the stomach uses depth-dependent mechanisms to respond to injury, leading to various histopathological changes and potential progression to dysplasia and cancer. This metaplasia process is broadly conserved across tissues and species, involving specific cytokines in facilitating the reprogramming of cells into different types, with implications for diagnostic and clinical purposes.

Profiling of the Bacterial Microbiota along the Murine Alimentary Tract

Ramiro Vilchez-Vargas et al.

Summary: This study examines the spatial distribution of bacterial flora in the murine alimentary tract and its correlation with Tff3 deficiency and colitis. The results show variations in bacterial species richness and community structure along different regions of the tract. Interestingly, while Tff3 deficiency is linked to increased colitis, it does not significantly affect the bacterial communities.

Self-Renewal and Cancers of the Gastric Epithelium: An Update and the Role of the Lectin TFF1 as an Antral Tumor Suppressor

Werner Hoffmann

Summary: Gastric cancer, predominantly caused by Helicobacter pylori infection, is the fourth leading cause of cancer deaths globally. It originates from dysregulated self-renewal of the gastric glands, which consist of two types of gastric units undergoing bi-directional self-renewal through stem cells.

Different Molecular Forms of TFF3 in the Human Respiratory Tract: Heterodimerization with IgG Fc Binding Protein (FCGBP) and Proteolytic Cleavage in Bronchial Secretions

Jens Weste et al.

Summary: Different molecular forms of TFF3 were identified in the lung using fast protein liquid chromatography and proteomics. The high-molecular mass form is TFF3-FCGBP oligomers, while the low-molecular mass forms are homodimeric and monomeric TFF3 with potentially anti-apoptotic activities. Disulfide-linked TFF3 heterodimers were also detected in bronchial secretions and BAL fluid. The study discusses a hypothetical model for how TFF3 may control FCGBP oligomerization and found no interactions between TFF3-FCGBP and DMBT1gp(340) or MUC5AC.

Spatiotemporal dynamics of self-organized branching in pancreas-derived organoids

S. Randriamanantsoa et al.

Summary: The development dynamics and self-organization of glandular branched epithelia is crucial for understanding tissue growth and cancer progression. Researchers successfully generated organoids from mouse pancreatic ductal adenocarcinoma cells that can mimic the complex architecture of PDAC. They identified distinct phases of morphogenesis and proposed a minimal theoretical model to capture the dynamics of tissue branching and proliferation. This study provides insights into the processes governing complex organoid structure formation and branching morphogenesis.

Trefoil Factor Family (TFF) Peptides and their Different Roles in the Mucosal Innate Immune Defense and More: An Update

Werner Hoffmann

Summary: Mucous epithelia are protected by complex mucus barrier layers, with Trefoil factor family peptides playing a key role in innate immune defense. TFF1 and TFF2 are mainly expressed in the gastric mucosa, while TFF3 is secreted widely from various mucous epithelia. Systematic analyses of the molecular forms of TFFs have revealed diverse functions, including lectin activities and interactions with proteins involved in mucosal innate immunity.

Trefoil Factor Family (TFF) Peptides and Their Links to Inflammation: A Re-evaluation and New Medical Perspectives

Werner Hoffmann

Summary: Trefoil factor family peptides (TFF1, TFF2, TFF3) and mucins are typical exocrine products of mucous epithelia with various physiological functions. TFF peptides, especially TFF2 secreted by myeloid cells and lymphocytes, play important roles in modulating inflammatory processes. These peptides have different lectin activities and may have potential implications in glycoimmunology research.
