Multi-marker profiling of extracellular vesicles using streaming current and sequential electrostatic labeling

Summary: Due to the high heterogeneity in membrane protein expression of small extracellular vesicles (sEVs), traditional antibody-based capture methods have limitations for expression analysis. In this study, an all-electric and microchip-based method was developed for unbiased analysis of sEV membrane protein expression and comparison of different sEV subsets. The method utilizes alternating charge labels to profile multiple surface proteins on captured sEVs. The results were validated using a fluorescence technique.

The potential use of mesenchymal stem cells-derived exosomes as microRNAs delivery systems in different diseases

Summary: MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression by targeting mRNA. Changes in miRNA expression have been observed in various diseases. Anti-miRNAs can suppress miRNA function, while modified miRNA mimetics can enhance miRNA function. Exosomes (EXOs), specifically those derived from mesenchymal stem cells (MSC-EXOs), have shown promise as miRNA carriers due to their therapeutic efficiency and safety. This article provides a summary of miRNAs and MSC-EXOs, including their production, purification, and miRNA loading methods, as well as the modification of MSC-EXOs for targeted miRNA delivery in different diseases.

Recent studies have explored the potential therapeutic element of microRNA (miRNA) in cancer treatment, with a focus on transferring miRNA to mesenchymal stem cells (MSC) and targeting miRNA-loaded exosomes to cancer cells. MSC-derived exosomes are gaining popularity as a non-cellular alternative to MSC-based therapy, as they avoid unwanted lineage differentiation.
