Somatic Mutation Profiling in Head and Neck Paragangliomas

Summary: This study analyzed somatic mutation profiles in HNPGLs and found that most patients had germline and somatic variants in paraganglioma susceptibility genes or related genes. The study also identified six significantly mutated genes and four mutational signatures associated with HNPGLs. These findings provide important insights into the molecular mechanisms involved in the development of HNPGLs.

Summary: Paragangliomas (PGLs) are rare neuroendocrine tumors that can develop from any paraganglion across the body. Carotid PGLs (CPGLs) are characterized by predominantly non-aggressive behavior; however, all tumors have the potential to metastasize. A case study of a 38-year-old woman with metastatic CPGL revealed a germline pathogenic splice acceptor variant in the SDHB gene, indicating the potential prognostic value of the TERT promoter mutation.
