Homoarginine in the cardiovascular system: Pathophysiology and recent developments

Vitali Koch et al.

Summary: Recent research has identified the endogenous non-proteinogenic amino acid L-homoarginine (L-hArg) as a novel biomarker and a direct contributor to cardiac dysfunction in cardiovascular disease. The association between low L-hArg levels and adverse cardiovascular events and mortality suggests that nutritional supplementation may be effective in improving cardiovascular health.

The Tryptophan and Kynurenine Pathway Involved in the Development of Immune-Related Diseases

Ai Tsuji et al.

Summary: The tryptophan and kynurenine pathway is important in the nervous, endocrine, and immune systems, as well as in the development of inflammatory diseases. Some kynurenine metabolites have anti-oxidative, anti-inflammatory, and/or neuroprotective properties. These metabolites may also possess immune-regulatory properties that could alleviate inflammation. Abnormal activation of the pathway may be involved in various immune-related diseases. Interestingly, kynurenine metabolites may impact brain memory and immunity through glial function modulation. Further understanding of this concept and the role of gut microbiota could lead to innovative treatments for immune-related diseases.

An integrated view of anti-inflammatory and antifibrotic targets for the treatment of NASH

Frank Tacke et al.

Summary: The development of treatments for non-alcoholic fatty liver disease and non-alcoholic steatohepatitis (NASH) has been challenging due to the complexity of the disease and the involvement of multiple pathways. Current approaches target metabolic dysregulation, inflammation, fibrosis, and immune responses. Potential therapies include nuclear receptor agonists, lipotoxicity modulators, genetic variant modification, hormonal agonists, microbiota interventions, and lifestyle changes. Inhibiting activation of hepatic stellate cells, immune cell modulation, and cell therapy are strategies to attenuate fibrosis. However, the heterogeneity of NASH poses a challenge in developing effective drugs for all patients.

Effects of Different Storage Conditions on Lipid Stability in Mice Tissue Homogenates

Erika Dorochow et al.

Summary: Lipids are crucial biomolecules with involvement in various physiological and pathological processes, making the elucidation of their presence in tissue samples highly important. However, tissue analysis presents challenges, as pre-analytical factors can significantly alter lipid concentration, affecting the reliability of research results. This study investigates the influence of pre-analytical factors on lipid profiles in homogenized tissues. The results suggest that swift processing of tissue homogenates under cooled conditions is a viable option for reliable lipid analysis.

Eosinophil-derived IL-4 is necessary to establish the inflammatory structure in innate inflammation

Anja Kolbinger et al.

Summary: This study investigates the development and maintenance of inflammatory microenvironments during Toll-like receptor 2-mediated paw inflammation. It is found that immune cells are organized into three zones, including a pathogen-containing core region, a proinflammatory region, and an anti-inflammatory region. Eosinophils present in all three regions adapt their cytokine profile and their depletion affects resolution of inflammation.

Pre-analytical sample handling standardization for reliable measurement of metabolites and lipids in LC-MS-based clinical research

A. Sens et al.

Summary: The disciplines of lipidomics and metabolomics have potential in discovering diagnostic biomarkers, but proper sample handling is crucial due to ex vivo distortions. We evaluated the effect of intermediate storage temperature and period on analyte concentrations in plasma samples using LC-MS, and made recommendations for sample-handling protocols. Certain analytes were found to be unstable, emphasizing the need for meticulous processing. These recommendations will improve the reliability and quality of samples for routine clinical diagnosis involving metabolites.

The Potential Role of Cellular Senescence in Non-Alcoholic Fatty Liver Disease

Cornelius Engelmann et al.

Summary: Cellular senescence plays a significant role in non-alcoholic fatty liver disease (NAFLD), but further research is needed to understand the specific mechanisms. Hepatocyte senescence may contribute to fat accumulation, fibrosis, and inflammation, while senescence in non-parenchymal liver cells could potentially reduce fibrosis. A comprehensive understanding of the interaction between NAFLD and cellular senescence is crucial for identifying novel therapeutic targets.

Hepatic stellate cells in physiology and pathology

Dakota R. Kamm et al.

Summary: Hepatic stellate cells (HSCs) play critical roles in the normal liver and in response to injury. They are activated during liver injury and produce extracellular matrix in liver fibrosis. In the absence of injury, HSCs are in a quiescent state and store retinoids or vitamin A-containing metabolites. They also enhance the inflammatory response and express growth factors crucial for liver development and regeneration. Recent studies have identified diverse phenotypic alterations and unique subpopulations of HSCs.

Behind the Wall-Compartment-Specific Neovascularisation during Post-Stroke Recovery in Mice

Anja Kolbinger et al.

Summary: This study used the tMCAO mouse model and MELC for sequential immunohistochemistry to investigate the spatio-temporal alterations in the ischemic core following stroke. The results showed that significant angiogenesis occurs in the ischemic core around 14 days post-stroke, accompanied by the recruitment of immune cells. Additionally, the astroglial scar was found to play a role in the post-stroke regeneration process by separating different compartments and influencing angiogenesis.

Indoleamine 2,3-dioxygenase mediates the therapeutic effects of adipose-derived stromal/stem cells in experimental periodontitis by modulating macrophages through the kynurenine-AhR-NRF2 pathway

Hanyue Li et al.

Summary: This study investigated the effects and underlying mechanism of adipose-derived stromal/stem cell (ASC) therapy in experimental periodontitis. The results showed that ASCs can alleviate periodontitis by modulating macrophage polarization through the IDO-dependent Kyn-AhR-NRF2 pathway.

Cellular Bioenergetics: Experimental Evidence for Alcohol-induced Adaptations

Liz Simon et al.

Summary: At-risk alcohol use has significant effects on multiple organ systems and causes global health burden. Alcohol interferes with bioenergetic pathways in tissues, leading to cellular injury. This review focuses on the alcohol-induced bioenergetic adaptations in metabolically active tissues, such as the liver, heart, skeletal muscle, pancreas, and brain.
