Innate immune deficiencies are associated with severity and poor prognosis in patients with COVID-19

Marine Peyneau et al.

Summary: This study investigated the correlation between circulating innate phagocytes and disease severity in COVID-19 patients. The results showed abnormal phenotype of neutrophils and monocytes, elevated concentrations of activation markers in plasma. Critical patients exhibited increased immature and immunosuppressive neutrophils, downregulated HLA-DR monocytes, which were closely associated with disease severity. Additionally, impaired antimicrobial functions in neutrophils and monocytes of critical patients were correlated with organ dysfunction, severe infections, and mortality.

Emerging therapies for glioblastoma: current state and future directions

Liang Rong et al.

Summary: Glioblastoma (GBM) is a common brain tumor with poor prognosis. Immunotherapy, including immune checkpoint blockade, CAR T cell therapy, oncolytic virotherapy, and vaccine therapy, has shown promising results in improving GBM outcomes. Techniques to overcome the blood-brain barrier for targeted delivery are also being tested. This article reviews the rationales for these therapies, potential novel agents, current status of trials, and discusses challenges and future perspectives in glioblastoma immuno-oncology.

Randomized Phase 3 Trial of Ruxolitinib for COVID-19-Associated Acute Respiratory Distress Syndrome

Lindsay Rein et al.

Summary: This study evaluated the safety and efficacy of the Janus kinase (JAK)1/JAK2 inhibitor ruxolitinib in COVID-19-associated acute respiratory distress syndrome requiring mechanical ventilation. The results showed that ruxolitinib did not significantly reduce the 28-day mortality rate compared to placebo. However, there were numerical improvements with ruxolitinib in secondary outcomes.

Adaptive immunity to SARS-CoV-2 infection: A systematic review

Marcos Jesse Abrahao Silva et al.

Summary: Through this study, we can gain a brief overview of the main immunological biomarkers and their function in specific cell types during SARS-CoV-2 infection. In critically ill patients, adaptive immunity is diverse, aberrantly compromised, and appears late. The T-cell response, in particular, is an essential component for immunological memory and should be addressed in vaccine formulation strategies.

Characterizing the immune responses of those who survived or succumbed to COVID-19: Can immunological signatures predict outcome?

Ramin Sami et al.

Summary: The study found that changes in frequencies of certain immune cells and levels of specific immune factors could serve as potential predictors of mortality in COVID-19 patients. NK cell frequencies were significantly increased in patients who died during treatment, while monocyte numbers were elevated in both recovered and deceased patients. Th2, Th17, and Treg cell percentages were notably lower in deceased cases compared to healthy individuals and those who recovered.

Targeting Folate Metabolism Is Selectively Cytotoxic to Glioma Stem Cells and Effectively Cooperates with Differentiation Therapy to Eliminate Tumor-Initiating Cells in Glioma Xenografts

Masashi Okada et al.

Summary: The study found that folate antagonists such as MTX and pemetrexed have selective cytotoxic effects on GSCs, suggesting that folate metabolism may be a promising target for anti-GSC therapy. Furthermore, the combination of cytotoxic and differentiation therapies may be a novel approach to effectively eliminate cancer stem cells.

Critical role of Syk-dependent STAT1 activation in innate antiviral immunity

Shasha Liu et al.

Summary: Research has shown that activation of STAT1 in early viral infection is not dependent on cytokines and JAKs, but mainly through Syk. Deletion of Syk profoundly impairs immediate innate immunity and suppresses the antiviral response.

Evaluation of connectivity map shows limited reproducibility in drug repositioning

Nathaniel Lim et al.

Summary: The study compared the performance of CMap 1 and CMap 2, finding a low success rate in CMap 2 when prioritizing queried compounds, mainly due to the low reproducibility of differential expression between CMaps and within each CMap. Additionally, DE strength was predictive of reproducibility, influenced by compound concentration and cell-line responsiveness.

Low type I interferon response in COVID-19 patients: Interferon response may be a potential treatment for COVID-19

Ahmed Abdulwahid Salman et al.

Summary: In COVID-19 patients, IRF3 gene expression is significantly upregulated, while IFN-beta expression only slightly increases. Similar to SARS-CoV and MERS, COVID-19 infection does not efficiently induce an IFN response to reduce the severity of the virus. The results suggest that IFN-beta or alpha may be used as potential treatments.
