Three-dimensional non-invasive brain imaging of ischemic stroke by integrated photoacoustic, ultrasound and angiographic tomography (PAUSAT)

Summary: We conducted a study on ischemic stroke using the PAUSAT system, which combines photoacoustic computed tomography (PACT), high-frequency ultrasound imaging, and acoustic angiographic tomography. PAUSAT enables three-dimensional imaging of brain morphology, blood perfusion, and blood oxygenation. By studying two common ischemic stroke models in mice, we identified spatially heterogeneous hemodynamic changes in blood perfusion and oxygenation, as well as quantified tissue infarct volume. The results showed that PAUSAT is a valuable non-invasive tool for longitudinal studies of neurological diseases at the whole-brain scale.

Ultrasound and microbubble-mediated delivery of miR-424-5p has a therapeutic effect in preeclampsia

Summary: The study aimed to investigate the influence of ultrasound/microbubble-mediated delivery of miR-424-5p on trophoblast cells and its underlying mechanism. The results showed that miR-424-5p was downregulated while AOC1 was upregulated in the placental tissues of patients with preeclampsia. Overexpression of miR-424-5p activated the Wnt/beta-catenin signaling pathway and promoted the proliferation, migration, and invasiveness of trophoblast cells. Ultrasound/microbubble-mediated delivery of miR-424-5p enhanced the transfection efficiency and further promoted the effects of miR-424-5p in trophoblast cells. It also alleviated experimental preeclampsia in a mouse model.

Summary: The application of biocompatible nanocarriers in medicine has provided several benefits over conventional treatment methods. Therapeutic ultrasound (TUS) has shown potential in both standalone anticancer treatment and targeted drug release. By combining nanomedicine and ultrasound as a smart drug delivery system (DDS), it is possible to enhance in situ drug delivery, improve access to impermeable tissues, and achieve targeted drug release. However, further research and clinical trials are needed to understand the physical mechanisms and interactions involved and to overcome the challenges in advancing TUS as an efficient and safe cancer treatment.

Evaluating the effects of radiation and acoustically-stimulated microbubble therapy in an in vivo breast cancer model

Summary: Ultrasound-stimulated microbubbles (USMB) can enhance the effects of radiation therapy (XRT) on tumors by causing localized vascular effects. This study investigated the optimization of acoustic parameters for combining USMB and XRT in breast cancer xenograft tumors. The results showed that a pressure of 570 kPa, 1-minute exposure, and 1% (v/v) microbubble concentration resulted in significant cell death, while higher ultrasound pressure and exposure duration were required for significant microvascular disruption. Additionally, introducing a 6-hour delay between USMB and XRT did not further improve the tumor response compared to immediate XRT delivery.

Aging and AD can affect the BBB's response to FUS, increasing BBBO volume, extending BBBC timeline, and increasing BBB permeability. This study provides quantitative data on the BBB's response to FUS in relation to age and AD, offering new considerations for FUS interventions in aged and AD populations.

MR-guided ultrasound-stimulated microbubble therapy enhances radiation-induced tumor response

Summary: High intensity focused ultrasound (HIFU) systems are used to cause tissue ablation or induced hyperthermia, and can be enhanced by the use of microbubble agents. This study investigated the effectiveness of reduced acoustic power and pressure on a commercial MR-integrated HIFU system in combination with radiation therapy for tumor treatment. The results showed that the combined therapy group exhibited statistically significant tumor cell death compared to the control group.

Phase-transition nanodroplets with immunomodulatory capabilities for potentiating mild magnetic hyperthermia to inhibit tumour proliferation and metastasis

Summary: In this study, a hybrid nanosystem (SPIOs + RPPs) was developed to enhance mild magnetic hyperthermia immunotherapy, effectively inhibiting tumor proliferation and metastasis while minimizing damage to normal tissues. The nanosystem utilized phase transition nanodroplets with immunomodulatory capabilities to lower the temperature threshold of magnetic hyperthermia and enhance immune stimulation. These findings provide a novel strategy for clinical translation of enhanced mild magnetic hyperthermia immunotherapy and ultrasound imaging.

On the magnetic nanoparticle injection strategy for hyperthermia treatment

Summary: In this study, a computational framework combining the lattice-Boltzmann method (LBM) and the particle-swarm optimization (PSO) algorithm was developed to find optimal strategies for magnetic nanoparticle (MNP) injection in hyperthermia-based cancer treatment. The results showed that multi-site injection strategies performed better than single-site injection strategies, and the more injection sites, the better the performance. The assessment of thermal dose considering both temperature and heat exposure time also indicated that the optimal multi-site injection strategies worked well for both tumor models.
