Isolation and characterization of Staphylococcus argenteus strains from retail foods and slaughterhouses in Japan

Yuki Wakabayashi et al.

Summary: This study investigated the prevalence of S. argenteus in retail fresh food and poultry slaughterhouses in Japan. The results showed that S. argenteus was primarily detected in chicken samples, indicating a potential source of contamination. Genetic analysis revealed a long-term presence of S. argenteus strains in one poultry slaughterhouse and their genetic connection with strains isolated from chicken meat. This is the first study to demonstrate the presence of S. argenteus in a food processing facility and highlight the possibility of bacterial contamination during food processing.

Top-Down Mass Spectrometry for Trace Level Quantification of Staphylococcal Enterotoxin A Variants

Donatien Lefebvre et al.

Summary: This study highlights the importance of improving the sensitivity of top-down mass spectrometry for the identification, differentiation, and absolute quantification of SEA sequence variants. By utilizing immunoaffinity enrichment, a protein internal standard, and optimized acquisition conditions, the top-down method demonstrated efficient identification and precise quantification of SEA variants, as well as providing a supplementary specificity criterion for quantification. The successful evaluation on enterotoxin-producing strains isolated during food poisoning outbreaks showcases the potential of top-down mass spectrometry in complementing traditional methods for toxin analysis.

Quantitative Determination of Staphylococcus aureus Enterotoxins Types A to I and Variants in Dairy Food Products by Multiplex Immuno-LC-MS/MS

Donatien Lefebvre et al.

Summary: The study introduced a new food testing protocol utilizing high-resolution mass spectrometry for simultaneous and quantitative determination of the eight most frequent Staphylococcal enterotoxins, including sequence variants. The assay was validated in dairy food products with a lower limit of quantification down to 0.1 ng/g, and successfully demonstrated quantification of SEs in real-life samples. The method's ability to detect diverse sequence variants highlights its potential for facilitating laboratory confirmation of positive samples in food poisoning outbreaks.

Genome Sequencing of a Historic Staphylococcus aureus Collection Reveals New Enterotoxin Genes and Sheds Light on the Evolution and Genomic Organization of This Key Virulence Gene Family

Jo Dicks et al.

Summary: In this study, a collection of historic Staphylococcus aureus strains was used to conduct a computational analysis of enterotoxin genes. Two novel staphylococcal enterotoxin genes were identified, along with additional gene variants. The study also revealed the prevalence of cooccurrence of enterotoxin genes and the importance of clonal complex membership in determining enterotoxin load.

Genomic Analysis of Global Staphylococcus argenteus Strains Reveals Distinct Lineages With Differing Virulence and Antibiotic Resistance Gene Content

Cosmika Goswami et al.

Summary: Infections due to Staphylococcus argenteus have been increasingly reported worldwide and the microbe cannot be distinguished from Staphylococcus aureus by standard methods. Its complement of virulence determinants and antibiotic resistance genes remain unclear, and how far these are distinct from those produced by S. aureus remains undetermined. Staphylococcus argenteus possesses important genetic differences from Staphylococcus aureus, with the potential to produce distinct clinical manifestations.

Phenotypic and Genomic Profiling of Staphylococcus argenteus in Canada and the United States and Recommendations for Clinical Result Reporting

AliReza Eshaghi et al.

Summary: This study describes the largest collection of Staphylococcus argenteus isolates in North America, emphasizing identification challenges and recommending whole-genome sequencing analysis for confirmation. The majority of isolates belonged to MLST ST2250/coa type XId with high genomic homology but showed diversity in spa types. Some isolates carried mecA and other resistance and virulence determinants commonly associated with Staphylococcus aureus. The findings suggest recommendations for clinical laboratories to deal with this emerging pathogen.

Development and validation of the Standard method EN ISO 19020-microbiology of the food chain - Horizontal method for the immunoenzymatic detection of staphylococcal enterotoxins in foodstuffs

Y. Nia et al.

Summary: A method for detecting staphylococcal enterotoxins in food has been developed, validated, and standardized under the CEN Mandate M/381 from the European Commission. The method involves an extraction procedure based on dialysis concentration followed by immunoenzymatic detection. Two commercially available immuno-enzymatic kits met the performance criteria in an interlaboratory study, leading to recognition as the European Union reference method for detecting staphylococcal enterotoxins in food.
