Comparison of Six Lytic Polysaccharide Monooxygenases from Thermothielavioides terrestris Shows That Functional Variation Underlies the Multiplicity of LPMO Genes in Filamentous Fungi

Monika Tolgo et al.

Summary: This study provides a detailed functional characterization of six AA9 LPMOs from Thermothielavioides terrestris LPH172. The research shows that these LPMOs have different activities on various cellulose substrates, further supporting the hypothesis that the multiplicity of LPMO genes is related to the complex structure of lignocellulosic biomass.

Summary: The present study evaluated the probiotic properties of Bacillus subtilis KC1 as a feed additive in poultry feed. The supplementation of Bacillus subtilis improved growth performance, heat-stress tolerance, resistance to Mycoplasma gallisepticum and Salmonella Pullorum challenge, as well as liver function and immune response of broilers challenged with Aflatoxin B1. These results suggest a tremendous potential of Bacillus subtilis KC1 as a feed additive in poultry feed.

Two C1-oxidizing AA9 lytic polysaccharide monooxygenases from Sordaria brevicollis differ in thermostability, activity, and synergy with cellulase

Xi Zhang et al.

Summary: This study identified two C1-oxidizing AA9 LPMOs, SbLPMO9A and SbLPMO9B, from Sordaria brevicollis, and explored their differences in terms of thermostability, reducing agent specificity, and synergy with cellulase. The two enzymes showed weak binding to cellulose, intolerance to hydrogen peroxide, and differing specificities and responsivities toward electron donors. SbLPMO9A exhibited better thermostability and boosting effect on enzymatic hydrolysis of delignified wheat straw, suggesting high potential application in biorefineries for lignocellulosic biomass.

Unraveling the roles of the reductant and free copper ions in LPMO kinetics

Anton A. Stepnov et al.

Summary: The study describes a novel two-domain cellulose-active family AA10 LPMO from a marine actinomycete, and investigates the effects of the reductant and copper ions on the LPMO reaction. Results show that ascorbate-driven LPMO reactions are extremely sensitive to very low amounts of free copper, whereas reactions driven by gallic acid are almost unaffected by the presence of free copper ions.

Summary: In this study, two novel glucanase genes, Cel5A-h38 and Cel5A-h49, were isolated and heterologously expressed from the metatranscriptome of the Hu sheep rumen microbiome. The enzymes displayed distinct substrate specificity, with Cel5A-h38 being active exclusively in the presence of mixed-linked glucans and Cel5A-h49 exhibiting a wider substrate spectrum. The results provided new insights into glucanase activity and established a foundation for the bioconversion of lignocellulosic biomass.

Secreted pectin monooxygenases drive plant infection by pathogenic oomycetes

Federico Sabbadin et al.

Summary: The discovery of copper-dependent lytic polysaccharide monooxygenases (LPMOs) in plant pathogenic oomycetes, particularly Phytophthora infestans, has revealed their crucial role in early infection stages by oxidatively cleaving pectin in the plant cell wall. Silencing LPMO-encoding genes inhibits potato infection by P. infestans, demonstrating their significance in host penetration. Identifying LPMOs as virulence factors in pathogenic oomycetes presents new opportunities in crop protection and food security.
