Deciphering the role of cytoplasmic domain of Channelrhodopsin in modulation of the interactome and SUMOylome of Chlamydomonas reinhardtii

Komal Sharma et al.

Summary: In this study, two important motifs, VXPX and LKNE, were identified at the C-terminus of ChR1. VXPX is a ciliary targeting sequence in animals, while LKNE is a well-known SUMOylation motif. The study showed that VMPS of ChR1 is essential for interaction with GTPase CrARL11 and revealed the presence of SUMO motifs in the C-terminus of putative ChR1s from green algae. Additionally, the study demonstrated the potential role of SUMOylation in modulating ChR1 protein in Chlamydomonas.

Gating and ion selectivity of Channelrhodopsins are critical for photo-activated orientation of Chlamydomonas as shown by in vivo point mutation

Olga Baidukova et al.

Summary: The authors manipulated the photocycle kinetics and ion selectivity of channelrhodopsin through point mutations, providing a deeper understanding of phototaxis in low light conditions. Their study also highlights the potential of the CRISPR-Cas9 technique for precise genetic modifications.

Ca2+ elevations disrupt interactions between intraflagellar transport and the flagella membrane in Chlamydomonas

Cecile Fort et al.

Summary: In Chlamydomonas, the movement of adhesive glycoproteins in flagella membrane for gliding motility is directed by transient interactions with intraflagellar transport (IFT) trains. Flagella Ca2+ elevations initiate the movement of paused retrograde IFT trains, while high-frequency Ca2+ elevations on highly adherent surfaces prevent the accumulation of these trains. Ca2+ disrupts the interaction between retrograde IFT trains and flagella membrane glycoproteins, allowing precise control of traction force and gliding motility.

L-Type Ca2+ Channel Regulation by Calmodulin and CaBP1

James B. Ames

Summary: CaM and CaBP1 interact with CaV through binding to the IQ motif to oppositely control Ca2+-dependent channel activity, with CaM promoting CDI and CaBP1 preventing CDI and promoting CDF.

Structural determinants of voltage-gating properties in calcium channels

Monica L. Fernandez-Quintero et al.

Summary: Voltage-gated calcium channels play a crucial role in controlling key functions of excitable cells. By analyzing the voltage sensing domains (VSDs), it was found that specific ion-pair formation in separate VSDs can realize the characteristic gating properties of voltage-gated cation channels. This mechanism provides a mechanistic model explaining how the stabilization of activated and resting states affects the kinetics and voltage dependence of activation.
