The findings have important implications for the development of HP-MOFs as drug carriers and controlled drug release systems.

Biomimetic Metal-Organic Frameworks as Targeted Vehicles to Enhance Osteogenesis

Summary: In this study, a biomimetic nanoplatform was developed using stem cell membrane-wrapped dexamethasone-loaded zeolitic imidazolate framework-8. The nanoplatform achieved efficient drug delivery and enhanced bone repair through its high loading capacity, sustained release, and specific uptake by mesenchymal stem cells. The nanoplatform showed high cytocompatibility, greatly enhanced osteogenic differentiation, and activated the PI3K-Akt signaling pathways involved in accelerated osteogenesis. In a bone defect model, the nanoplatform exhibited excellent biocompatibility and effectively promoted bone regeneration. This research provides a new and valid strategy for treating tissue injuries.

Enhanced Cancer Starvation Therapy Enabled by an Autophagy Inhibitors-Encapsulated Biomimetic ZIF-8 Nanodrug: Disrupting and Harnessing Dual Pro-Survival Autophagic Responses

Summary: In this study, a novel nanodrug system was designed to achieve an enhanced anticancer effect through an autophagy regulation strategy and starvation therapy. The results showed that ZIF-8 as a nanocarrier could induce autophagy for promoting cancer cell survival, while CQ could inhibit this autophagy and achieve the anticancer effect. Moreover, starvation caused by GOx could harness autophagy for maintaining intracellular ATP levels, and CQ could further inhibit autophagy and enhance the anticancer efficiency of starvation therapy.

Alendronate crosslinked chitosan/polycaprolactone scaffold for bone defects repairing

Summary: In this study, the researchers evaluated the osteogenic differentiation of ectomesenchymal stem cells (EMSCs) using an alendronate-loaded chitosan/polycaprolactone scaffold (CS/PCL@AL) in vitro. The results showed that the CS/PCL@AL scaffold enhanced osteogenic differentiation by upregulating bone morphogenetic protein 2, interleukin 10, and laminin expression, as well as activating the yes-associated protein (YAP) signaling pathway. In vivo experiments in rats with calvarial defects also demonstrated that the CS/PCL@AL scaffold promoted new and mature bone formation. This study highlights the potential of CS/PCL@AL scaffolds as an efficient delivery system for bone defect repair.

Structural resolution and mechanistic insight into hydrogen adsorption in flexible ZIF-7

Summary: Flexible metal-organic frameworks, such as ZIF-7, show potential for high usable hydrogen storage capacities with minimal swings in pressure and temperature. By conducting high-pressure isothermal hydrogen adsorption measurements, researchers were able to identify the pressure-temperature conditions of hydrogen-induced structural transition in ZIF-7, revealing narrow hysteresis and shallow adsorption slope between 100 K and 125 K. This study provides insights into the cause of phase transition in ZIF-7 and highlights the potential for future tuning of step-shaped adsorption in flexible porous adsorbents.
