Transgene-free genome editing supports the role of carotenoid cleavage dioxygenase 4 as a negative regulator of β-carotene in banana

Praveen Awasthi et al.

Summary: Carotenoid cleavage dioxygenases (CCDs) play an important role in the metabolism of carotenoids in plants. In this study, we investigated the regulatory mechanism of beta-carotene homeostasis in banana using transgene-free genome editing. We found that CCD4 is a key regulator of beta-carotene degradation and its expression is correlated with carotenoid concentrations. Additionally, our study demonstrated the successful use of genome editing in increasing beta-carotene accumulation in biofortified banana lines.

Metabolic engineering in food crops to enhance ascorbic acid production: crop biofortification perspectives for human health

Siddhant Chaturvedi et al.

Summary: Ascorbic acid (AsA), also known as vitamin C, plays important roles in human health and plant physiology. It is essential for various physiological processes and has antioxidant properties. AsA deficiency is a concern in both developed and developing countries. Biofortification of staple crops with AsA can be a feasible solution.

Analysis of TCP Transcription Factors Revealed Potential Roles in Plant Growth and Fusarium oxysporum f.sp. cubense Resistance in Banana (cv. Rasthali)

Siddhant Chaturvedi et al.

Summary: This study retrieved 27 non-redundant TCP gene sequences from the banana genome and analyzed their characteristics and phylogenetic relationships. The research found that these genes are highly conserved among different species and play an important role in banana growth, development, and Fusarium infection.

Over-expression of GGP1 and GPP genes enhances ascorbate content and nutritional quality of tomato

Athanasios Koukounaras et al.

Summary: This study aimed to enhance the AsA content in tomato fruit by overexpressing GDP-L-Galactose phosphorylase (GGP) and L-galactose-1-phosphate phosphatase (GPP). The transgenic lines showed increased levels of AsA without affecting fruit characteristics. RNA-Seq analysis revealed that the overexpression of GGP affected the expression of genes involved in cell wall modification, ethylene biosynthesis and signaling, pollen fertility, and carotenoid metabolism. This research provides valuable insights into the role of AsA induced genes in important traits, breeding programs, and gene editing approaches.

The role of GDP-L-galactose phosphorylase in the control of ascorbate biosynthesis

Mario Fenech et al.

Summary: The study analyzed the subcellular localization and potential interactions of enzymes involved in L-ascorbate biosynthesis pathway in photosynthetic organisms. It was found that most enzymes showed dual cytosolic/nuclear localization and could interact with each other through coimmunoprecipitation, although direct interactions were not observed in yeast-two hybrid analysis. The research highlighted the crucial role of GDP-L-galactose phosphorylase (GGP) in regulating ascorbate synthesis and its impact on enzyme interactions within the pathway.

Estimation of the Vitamin C Content and GDP-L-Galactose Phosphorylase Gene (VTC2) Expression Level in Leek (Allium porrum L.) Cultivars

O. K. Anisimova et al.

Summary: This study identified the expression patterns of the VTC2 gene and changes in AsA content in leek cultivars, finding variations in VTC2 expression levels and corresponding AsA content in different cultivars. Additionally, under cold stress conditions, the expression of the VTC2 gene had a significant impact on AsA synthesis in green leaves.
