Surface translocation of ACE2 and TMPRSS2 upon TLR4/7/8 activation is required for SARS-CoV-2 infection in circulating monocytes

Summary: It has been a topic of debate whether human peripheral blood cells can be infected by SARS-CoV-2, as immune cells do not express mRNA of both ACE2 and TMPRSS2. Here, the researchers demonstrate that resting primary monocytes contain abundant cytoplasmic ACE2 and TMPRSS2 proteins, and circulating exosomes contain significant ACE2 protein. Upon TLR4/7/8 stimulation, ACE2 quickly translocates to the monocyte cell surface independently of ACE2 transcription, while TMPRSS2 surface translocation occurs with elevated mRNA expression. TLR-stimulated monocytes expressing both ACE2 and TMPRSS2 on the cell surface are efficiently infected by SARS-CoV-2, but this infection can be mitigated by drugs and antibodies. The study also shows that ACE2 surface translocation in peripheral myeloid cells from severe COVID-19 patients correlates with hyperactivation and PD-L1 expression.

Protective Effects of Dapsone on Scopolamine-Induced Memory Impairment in Mice: Involvement of Nitric Oxide Pathway

Summary: This study aimed to evaluate the effects of the anti-inflammatory drug dapsone on scopolamine-induced memory impairment in mice and investigate the role of nitric oxide (NO) in this process. The results showed that dapsone improved memory acquisition in scopolamine-induced memory impairment and exerted its neuroprotective effects through modulation of NO levels.

In an experimental model of cirrhosis, inhibition of IDO with 1-MT improved cardiac dysfunction and reduced systemic inflammation. Chronic administration of 1-MT also attenuated hepatic inflammation, fibrosis, and ductular proliferation in rats with biliary cirrhosis.

Losartan ointment attenuates imiquimod-induced psoriasis-like inflammation

Summary: The study demonstrates that topical application of 1% Losartan ointment can significantly alleviate IMQ-induced psoriasis symptoms, including reducing erythema and scaling, and decreasing skin thickness and T-cell infiltration. Losartan alleviates psoriasis by reducing AT1R and IL-17a expression. This research suggests that AT1Rs could be a promising therapeutic target in psoriasis.
