Evolution of Embryo Implantation Was Enabled by the Origin of Decidual Stromal Cells in Eutherian Mammals

Arun R. Chavan et al.

Summary: Evolutionary mechanism of ancestral inflammatory response transformed into embryo implantation in the eutherian lineage was elucidated by comparative study of several mammalian species. Decidual stromal cells were proposed to mediate the suppression of detrimental modules in the inflammatory response. The results provide a mechanistic understanding of early stages in the evolution of eutherian pregnancy.

NRP1 and MMP9 are dual targets of RNA-binding protein QKI5 to alter VEGF-R/ NRP1 signalling in trophoblasts in preeclampsia

Xingyu Yang et al.

Summary: The study demonstrates that QKI5 plays a significant role in regulating trophoblast proliferation and migration by controlling NRP1 expression, and thus contributes to the pathogenesis of PE. QKI5, through interaction with NRP1 and MMP9, mediates cell growth and movement. Overexpression of QKI5 alleviated symptoms in a PE mouse model, indicating its potential as a target for treatment strategies in acute PE patients.

Miscarriage matters: the epidemiological, physical, psychological, and economic costs of early pregnancy loss

Siobhan Quenby et al.

Summary: Miscarriage, characterized by the loss of a pregnancy before viability, has a significant global impact with a high occurrence rate and various risk factors. It can result in both physical and psychological consequences for individuals, highlighting the importance of providing mental health support and specialized obstetric care for women who have experienced miscarriage.

Decidual CXCR4+CD56brightNK cells as a novel NK subset in maternal-foetal immune tolerance to alleviate early pregnancy failure

Yu Tao et al.

Summary: The CXCR4(+)CD56(bright)dNK subset plays an important immunomodulatory role during early pregnancy, promoting Th2 shift and assisting in the establishment of immune-tolerance. This new dNK cell subset may provide important insights into NK cell mechanisms in early pregnancy, and offer potential prognostic factors for the diagnosis and therapy of recurrent miscarriage.

Lili Du et al.

Summary: The research shows that abnormal cell composition and communication in the decidua during early pregnancy are associated with RSA, leading to stromal cell demise and pregnancy failure. These findings provide valuable insights for understanding the pathology of RSA and may help in preventing pregnancy loss.

Tim-3/CTLA-4 pathways regulate decidual immune cells-extravillous trophoblasts interaction by IL-4 and IL-10

Mengdie Li et al.

Summary: Establishing maternal-fetal tolerance and successful placentation are crucial for a successful pregnancy, as disruption of immune balance and inadequate placental perfusion can lead to pregnancy-related complications. Extravillous trophoblasts play a key role in regulating decidual immune cells for fetal tolerance. Blocking Tim-3 and CTLA-4 pathways can lead to abnormal interaction between decidual immune cells and extravillous trophoblasts, resulting in poor placental development and increased fetal loss. IL-4 and IL-10 treatment may rescue the adverse effects of targeting Tim-3 and CTLA-4 on pregnancy outcomes, suggesting them as potential therapeutic strategies to prevent pregnancy loss induced by checkpoint inhibition.

Cytokines, Hormones and Cellular Regulatory Mechanisms Favoring Successful Reproduction

Marie-Pierre Piccinni et al.

Summary: During pregnancy, the semi-allogeneic nature of the conceptus makes it vulnerable to attacks from the maternal immune system. Protective mechanisms, such as the effects of progesterone and PIBF on immune functions, work to support a healthy pregnancy outcome. The modulation of cytokine production by progesterone and dydrogesterone, as well as the induction of Treg cells, play crucial roles in maintaining a normal materno-foetal interaction and preventing pregnancy pathologies.
