Structural insights into the activation of chemokine receptor CXCR2

Kaiwen Liu et al.

Summary: The study reveals the binding modes between CXCL8 and CXCR2, CXCR2 and G protein, as well as the detailed binding pattern of the allosteric antagonist 00767013. Further analysis also uncovers the unique shallow-pocket activation mechanism of CXCR2 and suggests a potential role of cholesterol in allosteric modulation in chemokine receptors.

New Mutations and Pathogenesis of Pulmonary Hypertension: Progress and Puzzles in Disease Pathogenesis

Micheala A. Aldred et al.

Summary: Pulmonary arterial hypertension (PAH) is a complex disease characterized by alterations in the pulmonary circulation, leading to right heart failure and death. Mutations in the BMPRII gene account for a majority of PAH cases, but rare mutations in other genes have also been identified. Understanding these new PAH-related genes can provide insights into the pathogenesis and potential therapeutic targets for this incurable disorder.

Mechanistic insight into premature atherosclerosis and cardiovascular complications in systemic lupus erythematosus

Yudong Liu et al.

Summary: Systemic lupus erythematosus (SLE) is strongly associated with cardiovascular disease (CVD), and the interaction between immune dysregulation and traditional CVD risk factors plays a significant role in the development of premature atherosclerosis and CVD in SLE. Abnormal systemic inflammation in SLE can lead to lipid profile and function irregularities, further promoting a pro-atherosclerotic environment. Profound immune dysregulation, including excessive type I interferon burden, aberrant macrophage, platelet and complements activation, etc., contributes to accelerated CVD in SLE.

Single-Cell Analysis Reveals the Range of Transcriptional States of Circulating Human Neutrophils

Gustaf Wigerblad et al.

Summary: Neutrophils, previously thought to be homogeneous and transcriptionally inactive, have been found to be transcriptionally heterogeneous and can be classified into four clusters. This discovery, made possible by modifications to the data analysis pipeline, has potential implications for the characterization of neutrophils in health and disease.

Neutrophils Enhance Cutaneous Vascular Dilation and Permeability to Aggravate Psoriasis by Releasing Matrix Metallopeptidase 9

Jiaoling Chen et al.

Summary: Neutrophil infiltration and papillary vessel dilation are characteristic features of the initiation phase of psoriatic lesions. Our study found that neutrophils activate cutaneous vascular endothelial cells, leading to vascular remodeling, vasodilation, and enhanced vascular permeability both in vivo and in vitro. Furthermore, MMP-9 secreted by psoriatic neutrophils activates vascular endothelial cells and enhances CD4(+) T-cell transmigration, facilitating the development of psoriatic lesions.

Type I interferon activation and endothelial dysfunction in caveolin-1 insufficiency-associated pulmonary arterial hypertension

Salina Gairhe et al.

Summary: Interferonopathies, IFN-alpha/beta therapy, and CAV1 loss-of-function are associated with PAH. CAV1-silenced PAECs exhibit proliferative and hypermigratory behavior, with activation of STAT and PI3K/AKT pathways, leading to an IFN-biased inflammatory signature. Cav1(-/-) mice with pulmonary hypertension show STAT1 and AKT activation, increased CXCL10 levels, and PAH patients with CAV1 mutations display similar phenotypic features.

Summary: Neutrophils exhibit a single developmental spectrum, ranging from immature pre-neutrophils in the bone marrow to mature neutrophils in the blood and spleen. Inflammation affects neutrophil transcriptional states, leading to varying transcriptional activities at different sites.

Cell landscape atlas for patients with chronic thromboembolic pulmonary hypertension after pulmonary endarterectomy constructed using single-cell RNA sequencing

Ran Miao et al.

Summary: This study aimed to construct an atlas of the cell landscape and comprehensively characterize the cellular repertoire of pulmonary endarterectomized tissues in patients with CTEPH. Through single-cell RNA sequencing, the study identified 17 cell clusters classified into eight cell types, providing valuable insights into disease pathophysiology. Features of fibroblast/smooth muscle cells, endothelial cells, immune cells, and other cell types were significantly enriched with various functions, while CRISPLD2+ cell showed no significant enrichment, highlighting potential therapeutic targets.

Metalloproteinases and their inhibitors are associated with pulmonary arterial stiffness and ventricular function in pediatric pulmonary hypertension

Michal Schafer et al.

Summary: The disturbed balance between matrix metalloproteinases (MMPs) and tissue inhibitors (TIMPs) is recognized as a pathophysiological component of pulmonary arterial hypertension (PAH). This study found that levels of MMPs and TIMPs in children with PAH were associated with standard cardiac MRI hemodynamic indices and markers of pulmonary arterial stiffness, suggesting their potential as prognostic indicators in PAH.

Summary: The study revealed that PAH patients have elevated levels of NE and decreased levels of elafin, which are associated with more severe clinical disease and outcomes. NE levels can serve as a specific biomarker for the treatment of elafin.
