Sexually Dimorphic Neurotransmitter Release at the Neuromuscular Junction in Adult Caenorhabditis elegans

Summary: Sexual dimorphism in neurotransmitter release at the neuromuscular junctions (NMJs) of adult Caenorhabditis elegans has been observed. Adult males show enhanced neurotransmitter release and increased sensitivity to extracellular calcium concentration in their cholinergic synapses. Furthermore, male cholinergic synapses exhibit weaker synaptic depression but faster vesicle replenishment compared to hermaphrodites.

An Optogenetic Arrhythmia Model-Insertion of Several Catecholaminergic Polymorphic Ventricular Tachycardia Mutations Into Caenorhabditis elegans UNC-68 Disturbs Calstabin-Mediated Stabilization of the Ryanodine Receptor Homolog

Summary: In this study, six CPVT-causing mutations were inserted into the Caenorhabditis elegans homologs of RyR and CASQ, and their functional characteristics were studied using optogenetics and a microfluidic chip system. The results showed that three mutations reduced the organs' ability to respond to optogenetic stimulation, with two of them being reversible with a specific drug. This genetically amenable nematode model combined with optogenetics and high-throughput recordings provides a promising platform for investigating RyR mutations and selecting mutation-specific drugs.
