Polygenic risk provides biological validity for the ICHD-3 criteria among Finnish migraine families

Paavo Happola et al.

Summary: The study showed a correlation between the International Classification of Headache Disorders (ICHD-3) diagnostic criteria and the polygenic risk score of migraine, with the polygenic risk score associated with different types of migraine and their clinical presentations. This provides further biological support for the ICHD-3 diagnostic criteria.

Genome-wide analysis of 102,084 migraine cases identifies 123 risk loci and subtype-specific risk alleles

Heidi Hautakangas et al.

Summary: This study identified 123 susceptibility loci for migraine through genome-wide association analyses and implicated neurovascular mechanisms in its pathophysiology. Subtype analyses revealed risk loci specific for migraine with or without aura, as well as shared risk variants. The findings highlight the importance of both vascular and central nervous system tissue/cell types in the genetic underpinning of migraine.

Genome-wide association study reveals susceptibility loci for self-reported headache in a large community-based Asian population

Yu-Chien Tsao et al.

Summary: This study identified two novel loci, rs10493859 in TGFBR3 and rs13312779 in FGF23, associated with self-reported headache in Asian populations. Gene enrichment analysis indicated that these genes were significantly enriched in artery and adipose tissue. These results suggest that vascular dysfunction may play a role in the pathogenesis of self-reported headache in Asians.

Is periodontal disease a risk indicator for colorectal cancer? A systematic review and meta-analysis

Weiqi Li et al.

Summary: This study found a significant association between periodontal disease (PD) and colorectal cancer (CRC), with a 44% increased risk of CRC. Further research is needed to assess causality and effective periodontal treatment could be a preventive measure for CRC.

Increased Risk of Migraine in Patients with Chronic Periodontitis: A Population-Based Cohort Study

Yung-Kai Huang et al.

Summary: This study found an association between chronic periodontitis (CP) and an increased risk of subsequent migraine in Taiwan, with CP patients having a significantly higher incidence rate of migraine than non-CP patients during a 13-year follow-up period. Females also had a higher risk for migraine compared to males.

The Oral Microbiome in Periodontal Health

Magdalena Lenartova et al.

Summary: The estimation of oral microbiome (OM) taxonomic composition in periodontally healthy individuals can be biased, as the clinically periodontally healthy subjects may already have dysbiosis; By characterizing the OM in young individuals with good oral hygiene, distinct clusters corresponding to different health states were identified, with two possible oral types representing microbial succession.

Local and systemic mechanisms linking periodontal disease and inflammatory comorbidities

George Hajishengallis et al.

Summary: Periodontitis is causally linked to the development of other chronic inflammatory diseases outside the oral mucosa, and intervention studies have shown that local treatment of periodontitis can ameliorate surrogate markers of comorbid conditions. Experimental animal studies have further strengthened the potential causal link by establishing biologically plausible and clinically consistent mechanisms. Understanding the impact of disseminated periodontal pathogens and systemic inflammation associated with periodontitis on extra-oral pathologies may lead to new therapeutic options to reduce the risk of periodontitis-associated comorbidities.
