Mesenchymal Lineage Heterogeneity Underlies Nonredundant Functions of Pancreatic Cancer-Associated Fibroblasts

Erin J. Helms et al.

Summary: This study reveals the origins and functions of distinct cancer-associated fibroblast (CAF) subtypes in pancreatic ductal adenocarcinoma (PDAC). It shows that the abundant and transcriptionally diverse CAF population in PDAC arises from pancreatic stellate cells (PSC), but only a minor subset is derived from PSCs. These PSC-derived CAFs have unique functions in shaping the PDAC microenvironment.

Interleukin-17 activates and synergizes with the notch signaling pathway in the progression of pancreatic ductal adenocarcinoma

Xianze Wang et al.

Summary: The study confirmed the pro-tumor effects of IL-17 and Notch pathways in PanIN/PDAC progression and identified a positive correlation between IL-17 and Notch. Furthermore, co-inhibition of IL-17 and Notch pathways was shown to enhance therapeutic efficacy by restricting tumor growth.

IL17A critically shapes the transcriptional program of fibroblasts in pancreatic cancer and switches on their protumorigenic functions

Gianluca Mucciolo et al.

Summary: Interleukin-17A (IL17A) plays a crucial role in tumor fibroblasts and the tumor microenvironment, with IL17A knockout leading to increased desmoplastic reaction and altered immune cell infiltration. Fibroblasts with and without IL17A exhibit distinct genetic profiles, suggesting potential as a target for immune-based therapies in pancreatic ductal adenocarcinoma.

IFN-γ- and IL-17-producing CD8+ T (Tc17-1) cells in combination with poly-ICLC and peptide vaccine exhibit antiglioma activity

Takayuki Ohkuri et al.

Summary: While adoptive transfer of T-cells has been successful for B cell malignancies, developing effective T-cell-based immunotherapy for CNS tumors like glioblastoma remains challenging. Tc17-1 cells, which produce both IL-17 and IFN-gamma, showed promising results in combination with poly-ICLC and peptide vaccine treatment in a preclinical GBM model.

TNF-α-producing macrophages determine subtype identity and prognosis via AP1 enhancer reprogramming in pancreatic cancer

Mengyu Tu et al.

Summary: Large-scale genomic profiling of pancreatic cancer has identified two distinct subtypes - 'classical' and 'basal-like' - within the stromal immune microenvironment, influencing differential prognosis. Basal-like neoplastic state is sustained through BRD4-mediated cJUN/AP1 expression, inducing CCL2 to recruit TNF-alpha-secreting macrophages, which in turn forces classical neoplastic cells into an aggressive state via lineage reprogramming. Integration of various data sources revealed JUNB/AP1 (classical) and cJUN/AP1 (basal-like) driven regulation of PDAC subtypes.
