Dynamic evolution of Mhc haplotypes in cynomolgus macaques of different geographic origins

Natasja G. de Groot et al.

Summary: The Mhc genes of cynomolgus macaque exhibit high polymorphism and play a critical role in immune defense. Different geographic populations of cynomolgus macaques have specific combinations of Mhc class I and II haplotypes, suggesting dynamic evolution driven by recombination and selection under different pathogenic pressures.

The complete sequence of a human genome

Sergey Nurk et al.

Summary: The Telomere-to-Telomere (T2T) Consortium has presented a complete sequence of a human genome, T2T-CHM13, which covers the whole genome except for the Y chromosome. This new sequence includes gapless assemblies, error corrections in previous references, and nearly 200 million base pairs of additional sequence with gene predictions, including protein coding genes. The completion of important regions allows for further studies on genetic variations and functions.

Antigen presentation by MHC-E: a putative target for vaccination?

Linda Voogd et al.

Summary: HLA-E is a monomorphic human antigen presentation molecule that can be targeted to develop vaccines regardless of genetic diversity. Predictive models for HLA-E peptide binding have been improved, allowing for the discovery of more peptide binders. HLA-E is capable of binding structurally divergent peptides of both self and microbial origin.

KIR3DL05 and KIR3DS02 Recognition of a Nonclassical MHC Class I Molecule in the Rhesus Macaque Implicated in Pregnancy Success

Rachel E. Nicholas et al.

Summary: Understanding the interaction between rhesus macaque killer-cell Ig-like receptors (KIRs) and MHC class I ligands such as Mamu-AG is crucial for studying the role of natural killer (NK) cells in this species. The study reveals variation in KIR3DL05 and KIR3DS02 responses to different alleles of Mamu-AG, highlighting the impact of Mamu-AG polymorphisms on KIR recognition.

Assessment of safety and immunogenicity of MHC homozygous iPSC-derived CD34+hematopoietic progenitors in an NHP model

Saritha S. D'Souza et al.

Summary: The administration of ex vivo expanded somatic myeloid progenitors is explored as a way to accelerate myeloid recovery. Recent studies have shown the potential of hematopoietic progenitors derived from MHC homozygous iPSCs in treating cytopenia with good tolerance and safety in experimental animals.

Alternative splicing and genetic variation of mhc-e: implications for rhesus cytomegalovirus-based vaccines

Hayden Brochu et al.

Summary: RhCMV-based vaccination against SIV in rhesus macaques can effectively control SIV infection in approximately 55% of vaccinated monkeys by eliciting MHC-E-restricted CD8+ T cells. Analysis of the Mamu-E genomic sequences in rhesus macaques reveals gene duplication and different allele groups that are associated with the lack of vaccine protection.

Cytomegaloviral determinants of CD8+ T cell programming and RhCMV/SIV vaccine efficacy

Daniel Malouli et al.

Summary: The unconventional CD8(+) T cell responses induced by RhCMV/SIV vectors were found to be mediated by a genetic rearrangement in RhCMV, with MHC-E-restricted CD8(+) T cells playing a key role in the anti-SIV efficacy. Translation of these findings to humans may require deletion of genes inhibiting these responses from the HCMV/HIV vector.

Rapid Characterization of Complex Killer Cell Immunoglobulin-Like Receptor (KIR) Regions Using Cas9 Enrichment and Nanopore Sequencing

Jesse Bruijnesteijn et al.

Summary: Long-read sequencing significantly improves the quality and contiguity of genome assemblies, but requires deep sequencing coverage to overcome low nucleotide accuracy. Researchers combined a targeted enrichment protocol with long-read sequencing to efficiently annotate complex KIR gene regions, successfully resolving and phasing highly complex KIR haplotypes. This strategy eliminates PCR-induced amplification errors and allows for rapid characterization of large and complex multigenic regions, applicable in multiple species even without a reference genome.

Vasanthan Jayakumar et al.

Summary: This study performed de novo genome assembly of cynomolgus macaque and common marmoset, achieving high scaffold N50 lengths. The assembly of cynomolgus macaque outperformed all available assemblies in terms of contiguity, providing valuable resources for non-human primate models.
