Nutrient transporters: connecting cancer metabolism to therapeutic opportunities

Zeribe Chike Nwosu et al.

Summary: Cancer cells depend on specific extracellular nutrients for their metabolism and growth. SLC transporters play a crucial role in acquiring these nutrients and transferring intracellular nutrients. Understanding the function of SLC transporters in cancer could provide therapeutic targets for effective treatment.

Metabolomics of airways disease in cystic fibrosis

Joshua D. Chandler et al.

Summary: This article reviews recent literature to identify the most promising metabolomic findings of cystic fibrosis airways disease. Reproducible findings include increased levels of amino acids and small peptides, as well as changes in phospholipids and sphingolipids. Other commonly altered pathways include adenosine metabolism, polyamine synthesis, and oxidative stress. These findings have the potential to serve as biomarkers and therapeutic targets, but require reevaluation in the era of highly effective modulator therapies.

Influence of plasma halide, pseudohalide and nitrite ions on myeloperoxidase-mediated protein and extracellular matrix damage

Shuqi Xu et al.

Summary: Myeloperoxidase (MPO) generates the bactericidal oxidant hypochlorous acid (HOCl) to destroy pathogens, but it also causes tissue damage and disease. This study found that plasma levels of thiocyanate (SCN-) can modulate MPO-mediated damage, while nitrite (NO2-) alters the type of protein modification.

Escherichia coli RclA is a highly active hypothiocyanite reductase

Julia D. Meredith et al.

Summary: Hypothiocyanite and hypothiocyanous acid (OSCN-/HOSCN) are acids released by the innate immune system that can damage bacteria and proteins. Certain bacteria can evade the harmful effects of HOSCN, leading to severe diseases. In Escherichia coli, the flavoprotein RclA can efficiently reduce HOSCN and protect the bacteria. This suggests HOSCN may have antimicrobial activity in the gut.

The effects of neutrophil-generated hypochlorous acid and other hypohalous acids on host and pathogens

Agnes Ulfig et al.

Summary: Neutrophils play a crucial role in protecting the body against infections by killing pathogens through phagocytosis and the release of hypohalous acids. The formation of hypohalous acids by myeloperoxidase during the oxidative burst is a key mechanism in neutrophil antimicrobial activity. However, these acids can also impact bacterial survival strategies and modulate the immune response in the host system.

Role of myeloperoxidase and oxidant formation in the extracellular environment in inflammation-induced tissue damage

Clare L. Hawkins et al.

Summary: The heme peroxidase family, especially MPO, generates powerful oxidants for pathogen removal but also causes tissue damage and inflammatory diseases. MPO binds to extracellular biomolecules, leading to localized damage on specific sites and species, affecting cellular dysfunction and altered gene expression. MPO's interaction with damaged ECM components may explain its accumulation at sites of inflammation.
