Site-Specific Characterization of Heat-Induced Disulfide Rearrangement in Beta-Lactoglobulin by Liquid Chromatography-Mass Spectrometry

Summary: This study optimized a LC-MS-based proteomic approach to investigate disulfide rearrangement in beta-lactoglobulin under different heat treatments. The results showed non-native disulfide bonds forming at around 70°C and increased inter-molecular disulfide links at temperatures >= 80°C, correlating with protein aggregation as detected by size exclusion chromatography. Cys66 was identified as a key residue in heat-induced disulfide rearrangement and protein aggregation in beta-LG.

Selective and sensitive UHPLC-ESI-Orbitrap MS method to quantify protein oxidation markers

Summary: A targeted UHPLC-MS/MS isotopic dilution method was developed for simultaneous quantification of 18 different free and protein-bound aromatic amino acid oxidation products. The method demonstrated high selectivity without derivatization or ion pairing agents. Stable isotope-labelled internal standards were used to compensate errors due to matrix effects, and both acid and enzymatic hydrolyses were tested to obtain accurate quantification results for protein oxidation products in various matrices.

Reactions of plant polyphenols in foods: Impact of molecular structure

Summary: This study focuses on the reaction mechanisms of plant polyphenols with food components and how the choice of polyphenols with specific molecular structure can control undesired reactions in foods. Polyphenols with o-diphenol structure oxidize in foods and react fast with amino acid residues, potentially changing protein functional properties. Additionally, polyphenols with hydroxyl groups in meta-position tend to favor reactions with carbonyls.
