Mono-n-butyl phthalate regulates nuclear factor erythroid 2-related factor 2 and nuclear factor kappa B pathway in an ovalbumin-induced asthma mouse model

Quang Luu Quoc et al.

Summary: Recent studies have found that endocrine-disrupting chemicals play a significant role in the development of asthma, particularly eosinophilic asthma. However, the specific mechanism remains unclear. This study used mouse models to investigate the harmful effects of mono-n-butyl phthalate (MnBP) on airway epithelial cells, T cells, and eosinophils. The results showed that MnBP increased airway hyperresponsiveness, eosinophil and T cell counts, and T helper 2 cytokine release in mice with asthma. Furthermore, MnBP directly enhanced eosinophil activation.

Short-term high-dose gavage of hydroxychloroquine changes gut microbiota but not the intestinal integrity and immunological responses in mice

Zhi-Yuan Pan et al.

Summary: Hydroxychloroquine has significant effects on the gut microbiota of mice, reducing microbial diversity and altering the relative abundance of certain bacterial phyla. However, short-term high dose HCQ challenge does not affect intestinal integrity and immune responses in mice. Special attention should be paid to the effects of HCQ on intestinal microecology in future clinical use.

Hydroxychloroquine can impair tumor response to anti-PD1 in subcutaneous mouse models

Simon Wabitsch et al.

Summary: This study evaluated the impact of HCQ treatment on the efficacy of anti-PD1 tumor immunotherapy and found that it can lead to immunotherapy failure due to its immunosuppressive effects that offset both increased MHC-I expression by tumor cells and direct cytotoxicity.

S100A9 in adult asthmatic patients: a biomarker for neutrophilic asthma

Quang Luu Quoc et al.

Summary: S100A9 is a potential serum biomarker and therapeutic target for neutrophilic asthma. Patients with neutrophilic asthma had higher levels of S100A9 protein in their blood, which could be used as a potential biomarker and therapeutic target.

Serum free culture for the expansion and study of type 2 innate lymphoid cells

Pablo de Lucia Finkel et al.

Summary: This study demonstrates a methodology for culturing and expanding mouse type 2 innate lymphoid cells in serum-free medium supplemented with interleukins. The cells significantly expand from low to high concentrations within a week and can be sub-cultured in early or late phases of exponential growth. These findings provide a basis for expanding ILC2 populations for further study and potential applications under defined, serum-free conditions.

Airway monocyte modulation relates to tumour necrosis factor dysregulation in neutrophilic asthma

Natalie M. Niessen et al.

Summary: This study found increased levels of soluble TNF receptors in the sputum of patients with neutrophilic asthma, which may be linked to the increased recruitment of monocytes and neutrophils into the airways and subsequent receptor shedding. Monocytes also contribute to the altered inflammatory environment in neutrophilic asthma.

IL-1β augments TGF-β inducing epithelial-mesenchymal transition of epithelial cells and associates with poor pulmonary function improvement in neutrophilic asthmatics

Shengding Zhang et al.

Summary: Neutrophilic asthmatics (NA) show less improvement in lung function after treatment, which is associated with increased IL-1β levels in sputum. Additionally, IL-1β may enhance steroid-resistant EMT induced by TGF-β1 via the MAPK signaling pathway.
