Summary: Understanding the factors that alter drug pharmacokinetics is crucial for optimal disease treatment to prevent unexpected toxicity or treatment failure. In this study, the effects of 1,25-dihydroxyvitamin D3 (VDR ligand) on the metabolism, transport, and pharmacokinetics of indinavir were investigated. The results showed that treatment with 1,25(OH)2D3 upregulated the expression of CYP3A4 and MDR1, leading to increased indinavir metabolism and efflux ratio. This study suggests that VDR may play an important role in drug interactions and improving disease treatments.

Furthermore, this review discusses promising antagonists and their clinical applications in reversing chemoresistance.

A new drug-drug interaction-tilmicosin reduces the metabolism of enrofloxacin through CYP3A4

Summary: Monitoring in China has shown a decrease in single-pollutant residues exceeding the standard in food, but an increase in the coexistence of multiple low-level residual pollutants. The combination of enrofloxacin and tilmicosin is particularly concerning. This study evaluated the effects of this drug combination on metabolism and residues in vivo and in vitro. The results indicate that tilmicosin affects the metabolism of enrofloxacin and increases its residual concentrations in broilers, while also prolonging the elimination time. Thus, reducing the combined use of tilmicosin and enrofloxacin in veterinary drugs and feeds is necessary to ensure human and animal safety.
