Autophagy genes in biology and disease

Hayashi Yamamoto et al.

Summary: Macroautophagy and microautophagy are highly conserved cellular processes in eukaryotes that involve the degradation of cellular material in lysosomes. These processes are regulated by autophagy-related proteins and other molecules, with some shared components between the pathways. Recent advancements in technology have allowed for a better understanding of the functions of individual autophagy gene products and their implications in human disorders. Additionally, genome data has shed light on the evolution of autophagy genes and their origins in prokaryotes.

ATG9 vesicles comprise the seed membrane of mammalian autophagosomes

Taryn J. J. Olivas et al.

Summary: ATG9 vesicles serve as the membrane seed for mammalian autophagosomes, as shown by Olivas et al. using nanodisc technology. The integration of ATG9 with expanding autophagosome membranes demonstrates the importance of lipid transfer in autophagosome expansion. This work provides significant insights into the model of autophagosome formation and expands our understanding of the cellular process of autophagy.

Morphology of Phagophore Precursors by Correlative Light-Electron Microscopy

Sigurdur Runar Gudmundsson et al.

Summary: This study used live-cell imaging and electron microscopy techniques to provide high-resolution morphology of phagophore precursors in three dimensions. It revealed that phagophores are already closed or nearly closed into autophagosomes at the stage when the omegasome diameter is still large. The phagophore precursors emerged as bud-like highly curved membrane cisterns next to the endoplasmic reticulum with a small opening to the cytosol, and then opened to form more flat cisterns that elongate and curve to form crescent-shaped phagophores.

Wetting regulates autophagy of phase-separated compartments and the cytosol

Jaime Agudo-Canalejo et al.

Summary: This study explores how autophagosomes sequester droplets containing p62 protein in cells and demonstrates the formation of double-membrane, autophagosome-like vesicles on protein-free droplets through partial wetting in vitro. A minimal physical model shows that droplet surface tension supports the formation of membrane sheets. The interaction between the material properties of droplets and membrane sheets elucidates the mechanisms underlying droplet autophagy.

An N-terminal conserved region in human Atg3 couples membrane curvature sensitivity to conjugase activity during autophagy

Yansheng Ye et al.

Summary: The enzyme Atg3 is crucial for autophagosome formation, facilitating the connection of LC3 to PE lipids. The N-terminal conserved region of human Atg3 communicates information from the curvature-sensing domain to the active site, promoting spatial regulation of LC3-PE conjugation on target membranes.

ATG4 family proteins drive phagophore growth independently of the LC3/GABARAP lipidation system

Thanh Ngoc Nguyen et al.

Summary: The ATG4 family of proteins promote autophagosome formation independently of ATG8 family processing and protease activity, and also have a role in transporting ATG9A vesicles to mitochondria with proximity partners like LRBA. Additionally, ATG4s facilitate phagophore-ER contacts during lipid-transfer phase of autophagosome formation and act as deubiquitinating-like enzymes in disassembling ATG8-protein conjugates.

The evolution of autophagy proteins - diversification in eukaryotes and potential ancestors in prokaryotes

Sidi Zhang et al.

Summary: Autophagy is a degradative pathway for cytoplasmic constituents that is conserved across eukaryotes. ATG genes have undergone extensive multiplications and losses in different eukaryotic lineages, resulting in functional diversification and specialization. Bacteria and archaea do not possess an autophagy pathway, but they do harbor some remote homologs of Atg proteins, indicating recruitment of preexisting proteins during eukaryogenesis.

Membrane perturbation by lipidated Atg8 underlies autophagosome biogenesis

Tatsuro Maruyama et al.

Summary: Lipidation of Atg8 is crucial for autophagosome biogenesis, with two aromatic membrane-facing residues playing important roles in membrane morphology and autophagy regulation. These findings have implications for understanding the mechanisms underlying efficient autophagy in yeast and mammalian cells.
