Network Pharmacology and Molecular Docking Study of the Chinese Miao Medicine Sidaxue in the Treatment of Rheumatoid Arthritis

Ning Wu et al.

Summary: This study investigated the molecular mechanisms of Compound Sidaxue (SX) in treating rheumatoid arthritis (RA) using network pharmacology and in vivo experiments. The active components and key targets of SX were identified using network pharmacology, and their effects were validated in a rat model of RA. The results suggest that SX can alleviate RA symptoms by modulating the VEGF/ PI3K/AKT signaling pathway, TNF-alpha signaling pathway, and IL/NF-kappa B signaling pathway.

Targeting the IL-5 pathway in eosinophilic asthma: a comparison of mepolizumab to benralizumab in the reduction of peripheral eosinophil counts

Arian Ghassemian et al.

Summary: This study compared the efficacy of mepolizumab and benralizumab in reducing peripheral eosinophil counts in patients with severe eosinophilic asthma. While both biologics showed significant reduction in eosinophil counts, benralizumab was able to achieve undetectable eosinophil counts in a higher percentage of patients compared to mepolizumab.

Understanding the key issues in the treatment of uncontrolled persistent asthma with type 2 inflammation

William W. Busse et al.

Summary: Asthma is a complex respiratory disease with various phenotypes, characterized by 2 type inflammation involving both the innate and adaptive immune systems, which underpins chronic inflammation and comorbidities. Targeting key type 2 cytokines has shown efficacy in personalized treatment, utilizing biomarkers downstream of type 2 inflammation for diagnosis and monitoring.

A mouse allergic asthma model induced by shrimp tropomyosin

Lei Fang et al.

Summary: This study developed a murine model induced by shrimp tropomyosin (ST) instead of ovalbumin (OVA) to study asthma pathogenesis and evaluate therapeutic drugs. Sensitization with ST induced significant levels of serum total IgE and ST-specific IgE, leading to severe eosinophilic airway inflammation. Additionally, there was an increase in CD4(+) T lymphocytes and production of IL-4 and IL-5 in BALF, as well as airway hyperresponsiveness to methacholine.

Tranglutaminase 2 contributes to the asthmatic inflammation by modulating activation of alveolar macrophages

Hyun Seung Lee et al.

Summary: TG2 expression is increased in both asthmatic patients and OVA-induced murine asthma model. TG2 deficiency reduces airway hyperresponsiveness and inflammatory cells in asthma mice, and normalizes the expression of M2 markers. In vitro experiments show that TG2 inhibitor treatment can attenuate IL-4 or IL-13-stimulated expression of M2 macrophage markers.

Macrophages: The Good, the Bad, and the Gluttony

Ewan R. A. Ross et al.

Summary: Macrophages play critical roles in sterile inflammation and autoimmune diseases, particularly in the pathophysiology of diseases like rheumatoid arthritis. Imbalance in macrophage homeostasis contributes to chronic inflammation, and therapeutic strategies targeting macrophage plasticity are being explored. Apoptotic cell-derived extracellular vesicles may play a role in regulating macrophage phenotype and are a potential target for new therapeutic approaches.

PLIP 2021: expanding the scope of the protein-ligand interaction profiler to DNA and RNA

Melissa F. Adasme et al.

Summary: With the growth of protein structure data, the analysis of molecular interactions between ligands and their target molecules becomes increasingly important. PLIP, a tool for protein-ligand interaction profiling, has been successful in detecting and visualizing these interactions, with applications ranging from characterizing docking experiments to evaluating novel ligand-protein complexes. Additionally, PLIP has been extended to encompass interactions with DNA and RNA, demonstrating its utility in scenarios such as cancer drug targeting DNA and RNA-protein complexes associated with neurological diseases.
