Enhancement of optical properties of new purine nucleobases containing electron-donating and -withdrawing peripheral groups

Summary: Currently, there is a growing interest in synthesizing highly fluorescent unnatural base analogs to overcome the lack of fluorescence in natural nucleic acid bases. These fluorescent unnatural base analogs, such as purines, can be used in various applications, including DNA and RNA optical spectroscopy studies. Additionally, the optical properties of purines base analogs, such as emission, can be adjusted through molecular engineering, making them more efficient as fluorescent probes. A new set of purines base analogs has been synthesized and studied for its optical spectroscopic properties. Molecules with NH-pi-CN arrangement showed an increase in fluorescence quantum yield, and lower energy 2PA state exhibited higher two-photon absorption (2PA) cross-sections. The enhancement of both properties makes these purines base analogs promising candidates for fluorescent probes in RNA and DNA spectroscopic studies.
