Regioselective C6-H Hydroxyalkylation of Purines and Purine Nucleosides via a-C-H Functionalization of Alcohols at Room Temperature

Summary: In this study, a highly regioselective C-6-H hydroxylalkylation method for purines and purine nucleosides was reported for the first time, which was achieved via the alpha-C(sp(3))-H functionalization of alcohols at room temperature within 10 minutes. The reaction displayed tolerance towards various functional groups and had the potential for further modification to yield valuable molecules. Compared to other methods, this strategy provided a direct approach to access 6-substituted alkylated purines and nucleosides with pharmaceutical bioactivities, without the need for metal catalysts, light, and protecting groups.

Traceless proton aided regioselective C(sp2)-C(sp2) construction to synthesize C6-acylated purines and purine nucleosides without metal catalysts

Summary: A highly regioselective radical reaction, aided by traceless H+, has been developed for the synthesis of C-6-acylated purines and purine nucleosides. This reaction is performed at room temperature within a short time period of 10 minutes. It offers excellent functional group compatibility, a broad substrate scope, and good C-6-regioselectivity. The use of H+ is environmentally friendly and does not require additional functional group modification.

Acid-base properties of an antivirally active acyclic nucleoside phosphonate: (S)-9-[3-hydroxy-2-(phosphonomethoxy)propyl]adenine (HPMPA)

Summary: HPMPA is an antiviral substance with interesting acid-base behavior. By measuring the chemical shifts of the protons in HPMPA, the protonation sites and acidity constants were determined. The phosphonate group is the most basic site, followed by the N1 position of adenine. The basicity of the adenine nitrogens decreases in the order N1 > N7 > N3.
