Anion-π Catalysis Enabled by the Mechanical Bond

Summary: We have reported a series of anion-it catalysts based on rotaxanes. The mechanical bond between a bipyridine macrocycle and an axle containing an NDI unit plays an intrinsic role in the observed activity. Our results show that a [3]rotaxane catalyst can selectively catalyze a Michael addition reaction, which is otherwise disfavored under Bronsted base conditions dominated by a decarboxylation pathway.

Summary: Cells process information similar to Turing machines, reading data from molecular tapes and translating it into outputs. However, synthetic small-molecule machines that can read and write information stored on artificial molecular tapes are still elusive. This study introduces a molecular ratchet system where a crown ether is transported onto an encoded molecular strand by a chemical fuel pulse. The crown ether is then transported through the tape compartments and changes conformation based on the binding sites, allowing the stereochemical information on the tape to be read out as a string of digits.

Rotaxanes as Cages to Control DNA Binding, Cytotoxicity, and Cellular Uptake of a Small Molecule**

Timothy Kench et al.

Summary: The efficacy of many drugs can be limited by undesirable properties, such as poor aqueous solubility, low bioavailability, and off-target interactions. Various drug carriers have been investigated to enhance the pharmacological profile of therapeutic agents. In this work, mechanical protection is used to cage a DNA-targeting metallodrug within a photodegradable rotaxane, which showed enhanced cell permeability and limited cytotoxicity within osteosarcoma cells compared to the free drug. Light activation led to a dramatic increase in cytotoxicity, arising from the translocation of Pt-II-salphen to the nucleus and its binding to DNA.

Cyclization of interlocked fumaramides into β-lactams: experimental and computational mechanistic assessment of the key intercomponent proton transfer and the stereocontrolling active pocket

Summary: This study investigates the CsOH-promoted diastereoselective cyclization of interlocked fumaramides to form beta-lactams. Experimental evaluation, KIE studies, and computational analysis indicate that the rate-determining step involves deprotonation of the N-benzyl group by the amidate group from the macrocycle. The polyamide macrocycle plays a crucial role in both activation and diastereoselection. Additionally, increased flexibility of the adamantyl core accelerates the cyclization process in diadamantyl-derived rotaxanes.
