Boosting Endogenous Copper(I) for Biologically Safe and Efficient Bioorthogonal Catalysis via Self-Adaptive Metal-Organic Frameworks

Summary: In order to boost the endogenous Cu(I) level for intracellular drug synthesis through the bioorthogonal reaction, a self-adaptive bioorthogonal catalysis system was constructed by encapsulating prodrugs and sodium ascorbate within adenosine triphosphate aptamer-functionalized metal-organic framework nanoparticles. The system presents specificity to tumor cells and does not require exogenous Cu catalysts, thereby leading to high anti-tumor efficacy and minimal side effects both in vitro and in vivo. This work will open up a new opportunity for developing biosafe and high-performance bioorthogonal catalysis systems.

Design, synthesis and biological evaluation of KRASG12C-PROTACs

Summary: Several small-molecule covalent inhibitors of KRASG12C have shown promising breakthroughs in treating KRAS mutant cancer. However, adaptive resistance may limit the clinical application of these inhibitors. The emerging PROTAC strategy offers complementary advantages and improves anti-tumor efficacy. Through protein degradation and anti-proliferative activities, novel KRASG12C-PROTACs have been demonstrated to effectively bind and degrade KRASG12C, surpassing the potency of AMG-510. Mechanistic studies indicate degradation effects through the ubiquitin-proteasome pathway. These findings support the use of PROTACs for degrading the oncogene KRASG12C and provide insights for optimizing the structure of KRASG12C-PROTACs.

Additionally, it demonstrated more anti-angiogenesis ability than sunitinib in both in vitro and in vivo studies.
