Matricellular Protein SPARCL1 Regulates Blood Vessel Integrity and Antagonizes Inflammatory Bowel Disease

Daniela Regensburger et al.

Summary: This study reveals that SPARCL1 inhibits angiogenesis, supports vessel morphogenesis and integrity, and has anti-inflammatory properties. The acidic domain of SPARCL1 is identified as necessary for its anti-angiogenic activity.

SVEP1 is a human coronary artery disease locus that promotes atherosclerosis

In-Hyuk Jung et al.

Summary: SVEP1 is a low-frequency variant associated with coronary disease risk and promotes atherosclerosis by affecting VSMC function and inducing inflammation to drive atherosclerosis. Pharmacological inhibition of SVEP1 may protect against atherosclerosis in humans.

Cardiovascular Events with Finerenone in Kidney Disease and Type 2 Diabetes

Bertram Pitt et al.

Summary: Finerenone therapy improved cardiovascular outcomes in patients with type 2 diabetes and stage 2 to 4 CKD with moderately elevated albuminuria or stage 1 or 2 CKD with severely elevated albuminuria, compared to placebo. The risk of the primary outcome event was significantly lower in the finerenone group, primarily driven by a reduction in hospitalization for heart failure.

Sparcl1 promotes nonalcoholic steatohepatitis progression in mice through upregulation of CCL2

Bin Liu et al.

Summary: This study identified increased expression of Sparcl1 in WAT from NASH mice and patients, positively correlated with hepatic pathological features. Sparcl1 promoted CCL2 expression in hepatocytes through TLR4 and NF-KB/p65 pathway, leading to hepatic inflammatory response. Blocking CCL2/CCR2 pathway attenuated the inflammatory response evoked by Sparcl1, suggesting Sparcl1 as a potential therapeutic target for NASH.

Large-scale plasma proteomic analysis identifies proteins and pathways associated with dementia risk

Keenan A. Walker et al.

Summary: The study identified 38 candidate proteins in nondemented older adults associated with future dementia risk. Pathway analysis of these proteins suggests immune, lipid, metabolic signaling, and hemostasis pathways play significant roles in dementia pathogenesis.
