A molecular device for the redox quality control of GroEL/ES substrates

Summary: Hsp60 chaperonins and their Hsp10 cofactors facilitate protein folding. This study reveals that the bacterial chaperonin GroEL forms a stable complex with the chaperedoxin CnoX, and CnoX likely functions as a redox quality-control plugin for GroEL. The findings suggest that Hsp60 molecular plugins have been conserved through evolution, as proteins sharing structural features with CnoX exist in eukaryotes.

Genetic design of co-expressing a novel aconitase with cis-aconitate decarboxylase and chaperone GroELS for high-level itaconic acid production

Summary: In this study, a new aconitase from acidophilic E. coli Nissle 1917, denoted as ENAcnA, was compared to Corynebacterium glutamicum for the first time. Co-expression of ENAcnA and cis-aconitate decarboxylase from Aspergillus terreus in recom-binant E. coli BL21(DE3) resulted in high-level IA production. Integration of GroELS chaperon into the chromosome of BL21(DE3) and cold treatment of the whole-cell biocatalyst further enhanced IA production.

Effective whole cell biotransformation of arginine to a four-carbon diamine putrescine using engineered Escherichia coli

In this study, a time-efficient method was developed to convert L-arginine into putrescine using whole cell bioconversion. Acetate-based production of itaconic acid with Corynebacterium glutamicum using an integrated pH-coupled feeding control

Summary: To date, most bio-based products of industrial biotechnology are derived from sugar-based carbon sources, which compete with food and feed resources. In this study, an engineered strain of Corynebacterium glutamicum ATCC 13032 was used to produce itaconic acid from acetate, overcoming current production challenges. The results show the potential of using acetate as an alternative substrate for bioproduction, with a high peak volumetric productivity and itaconate titer achieved.

Tailoring key enzymes for renewable and high-level itaconic acid production using genetic Escherichia coli via whole-cell bioconversion

Summary: Renewable chemical production through carbon-neutral design has received significant attention. In this study, the WC bioconversion of citrate into IA using key enzymes ACN and CAD showed promising results, with CAD identified as the rate-determining step. Metal ions like copper and ferric ions were found to inhibit enzyme activity when both enzymes co-worked. The engineered E. coli expressing dual genes achieved a high IA titer and productivity, demonstrating a promising renewable process.

Summary: In this study, the impact of Lac operon on protein expression in the T7-derived EcN strains was investigated. Through fine-tuning different parameters, the conversion rate of p-coumaric acid, a bioactive compound beneficial for human health, was significantly improved in simulated gut conditions. These findings highlight the potential of using EcN as an efficient cell factory for live bacterial therapeutics.

Summary: Engineering efficient itaconate producers often requires elimination of isocitrate dehydrogenase to preserve cis-aconitate, leading to auxotrophy for 2-ketoglutarate. This study presents a novel approach by introducing the Weimberg pathway for 2-ketoglutarate biosynthesis, overcoming the need for glutamate supplementation. Through careful tuning of xylose concentration, the final strain produced 20 g/L of itaconate in minimal medium without the need for antibiotics.
